不同采收期水冬瓜叶HPLC指纹图谱研究及质量标志物预测分析
HPLC fingerprint study and quality marker prediction analysis of the leaves of Toricellia Angulata Oliv.var.intermedia(Harms) Hu in different collecting seasons
【摘要】 建立不同采收期24批次水冬瓜叶药材的高效液相色谱(HPLC)指纹图谱,并采用化学模式识别法对影响不同采收期水冬瓜叶药材的质量标志物进行预测,为不同采收期水冬瓜叶质量控制及质量标志物预测提供方法和依据。采用Agela Promosil C18(250 mm×4.6 mm,5μm)色谱柱,以0.2%(体积分数)磷酸水溶液(A)-乙腈(B)为流动相,梯度洗脱,流量为1.0 mL/min,柱温为35℃,检测波长210 nm,进样量10μL。建立了不同采收期24批次水冬瓜叶HPLC指纹图谱,除Y19、Y23样本外,其余样本相似度为0.853~0.973。PCA将不同采收期24批次水冬瓜叶聚成3类。依据VIP>1,预测了影响不同采收期24批次水冬瓜叶药材的9个质量标志物,分别为峰14、峰15、峰11、峰12、峰7(金丝桃苷)、峰16、峰8(异槲皮苷)、峰9和峰17。建立的HPLC指纹图谱方法稳定、简便,主成分分析(principal component analysis, PCA)和正交偏最小二乘判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA)预测的影响不同采收期水冬瓜叶药材的质量标志物可为水冬瓜叶药材质量控制提供参考。
【Abstract】 The fingerprints of 24 batches of the leaves of Toricellia Angulata Oliv.var.intermedia(Harms) Hu in different collecting seasons were established by high performance liquid chromatography, and the quality markers of the leaves in different collecting seasons were predicted by the chemical pattern recognition method. This study provides a method and basis for quality control and quality marker prediction of the leaves in different collecting seasons. HPLC analysis was performed on a Agela Promosil C18 column(250 mm×4.6 mm, 5 μm). The mobile phase were consisted of 0.2% phosphoric acid solution(A) and acetonitrile(B)with gradient elution. The flow rate was 1.0 mL/min, the detection wavelength was 210 nm, the column temperature was 35 ℃ and the injection volumn was 10 μL. HPLC fingerprints of 24 batches of the in different collecting seasons were established. Despite the samples of Y19 and Y23, the similarity was 0.853~0.973. The24 batches of the leaves in different collecting seasons were clustered into three groups by PCA. According to VIP > 1, nine quality markers affecting 24 batches of the leaves of Toricellia Angulata Oliv.var.intermedia(Harms) Hu in different collecting seasons were predicted, which were peak 14, peak 15, peak 11, peak 12,peak 7(hyperin), peak 16, peak 8(isoquercitrin), peak 9 and peak 17, respectively. The established HPLC fingerprint method was stable and simple. The prediction of PCA and OPLS-DA could provide reference for the quality control of the leaves of Toricellia Angulata Oliv.var.intermedia(Harms) Hu in different collecting seasons.
