PRC1通过激活Wnt/β-catenin途径促进胶质母细胞瘤细胞增殖、迁移、侵袭和血管生成
PRC1 may promote the proliferation,migration,invasion and angiogenesis of glioblastoma cells through the activation of Wnt/β-catenin pathway
【摘要】 目的:探讨胞质分裂原蛋白调节因子-1(protein regulator of cytokinesis-1,PRC1)对胶质母细胞瘤的作用及其可能的机制。方法:RT-PCR和Western blot检测胶质母细胞瘤组织和正常脑组织中PRC1的表达;人胶质母细胞瘤细胞株U251转染siRNA-NC和siRNA-PRC1序列,RT-PCR检测细胞中PRC1 mRNA表达;Western blot检测细胞中PRC1、Wnt1、β-catenin和CyclinD1蛋白表达;CCK-8检测细胞增殖能力;流式细胞术检测细胞周期分布;Transwell实验检测细胞迁移和侵袭;血管生成实验检测血管生成。结果:与正常组织相比,癌组织中PRC1 mRNA和蛋白表达升高(P<0.001);与siRNA-NC组相比,siRNA-PRC1组细胞在24 h、48 h及72 h的细胞增殖能力降低(P<0.01),G0/G1期细胞含量升高(P<0.05),S和G2/M期细胞含量降低(P<0.05),细胞迁移数和侵袭数降低(P<0.01),内皮细胞管道形成数降低(P<0.001),Wnt1、β-catenin和CyclinD1蛋白表达降低(P<0.01)。结论:PRC1在胶质母细胞瘤组织中表达上调,PRC1可能通过促进Wnt/β-catenin途径活化,促进胶质母细胞瘤细胞增殖、细胞周期进程、迁移、侵袭和血管生成。
【Abstract】 Objective:To investigate the effect of protein regulator of cytokinesis-1(PRC1)on glioblastoma and its possible mechanism.Methods:RT-PCR and Western blot were used to detect the expression of PRC1 in glioblastoma tissue and normal brain tissue.Human glioblastoma cell line U251 was transfected with siRNA-NC and siRNA-PRC1 sequences,and RT-PCR was used to detect mRNA expression of PRC1 in cells.Western blot was used to detect protein expression of PRC1,Wnt1,β-catenin and CyclinD1 in cells.CCK-8 was used to detect cell proliferation ability.Flow cytometry was used to detect cell cycle distribution.Transwell experiment was used to detect migration and invasion of cells.Angiogenesis test was used to detect angiogenesis.Results:Compared with normal tissues,PRC1 mRNA and protein expression in cancer tissues were increased(P<0.001).Compared with siRNA-NC group,cell proliferation at 24 h,48 h and 72 h in siRNA-PRC1 group had decreased(P<0.01),G0/G1phase cell content increased(P<0.05),cell content in S and G2/M phase decreased(P<0.05),migration and invasion number of cells decreased(P<0.01),the number of tubes formed of endothelial cells decreased(P<0.001),and the expression of Wnt1,β-catenin and CyclinD1 protein decreased(P<0.01).Conclusion:The expression of PRC1is up-regulated in glioblastoma tissues.PRC1 may promote the proliferation,cell cycle progression,migration,invasion and angiogenesis of glioblastoma cells through the activation of Wnt/β-catenin pathway.
