磷脂酶D水解活性测定方法的研究
Methods for Determining Hydrolysis Activity of Phospholipase D
【摘要】 为解决磷脂酶D酶活测定试验重复性差、成本高等问题,研究并改良测定磷脂酶D水解活性的方法。将反应体系分为解离和显色两个过程,通过对反应底物、反应条件的优化试验,确定最佳底物为蛋黄来源的卵磷脂,底物溶剂为80%乙醇,其中解离过程的最佳反应条件为温度37℃、pH5.5、时间10 min,金属离子助剂为10 mmol/L的CaCl2。
【Abstract】 Due to the poor reproducibility and high cost of the traditional methods,this study aims to improve the method for determining the hydrolysis activity of phospholipase D. The reaction system was divided into two processes: dissociation and color development. The reaction substrate and reaction conditions were then optimized.The results demonstrated that the optimal substrate and solvent were egglecithin and 80% ethanol,respectively.The optimal conditions for the dissociation process were 37 ℃,pH5.5,10 min,and 10 mmol/L CaCl2.
