枇杷叶中熊果酸的提取工艺及含量测定研究

【摘要】 为探索枇杷叶中熊果酸的提取工艺和含量测定方法。以干燥的枇杷叶为原料,考察指标设为枇杷叶中熊果酸的含量,采用超声波提取法提取样品有效成分,分别考察提取溶剂、溶剂用量、以及提取次数与提取过程所用时间对提取量造成的影响。结果表明:枇杷叶中提取熊果酸效率最高的溶剂是甲醇溶液,溶剂用量为45 mL,常规提取过程所用时间是40 min,提取次数2次,甲醇-水-磷酸(90∶10∶0.05)为流动相。在上述条件下熊果酸保留时间短,基线平稳,峰形对称且无干扰,出峰情况良好,熊果酸提取率最高,含量达到0.798%。同时,用这种方法还有很多其他好处,操作过程较为简单,提取时间较少,重现性好,提取效果高,完全可以作为控制医学领域枇杷叶药材质量的一个方法。

【Abstract】 To explore the extraction process of ursolic acid from loquat leaves and methods for determining its content.With dried loquat leaves as raw material, ultrasonic extraction sample solution was used to examine effects of different extraction solvent, extracting times, extraction time of extraction on the extraction amounts.The most effecient solvent for extracting the ursolic acid was methanol solution, and solvent consumption was 45 mL, extracting time was 40 min, extraction times was 2 times, methanol-water-phosphoric acid(90∶10∶0.05) was taken as mobile phase. Under the above conditions, retention time of ursolic acid was short, baseline was smooth, peak form was symmetric and there was no interference, the peak was in good condition, ursolic acid extraction yield was highest with content of 0.798%.This method is simple, sensitive, fast and has good reproducibility, and can be used for the quality control of the medicinal material loquat leaves.
