非小细胞肺癌组织中miR-130a、GOLPH3表达变化及意义
Expression and clinical significance of miR-130a and GOLPH3 in non-small-cell lung cancer tissues
【摘要】 目的 探讨非小细胞肺癌(NSCLC)癌组织中微小RNA(miR)-130a、高尔基磷蛋白3(GOLPH3)表达及临床意义。方法 选取87例NSCLC患者为研究对象,应用荧光定量PCR检测NSCLC癌和癌旁组织中miR-130a、GOLPH3 mRNA表达;采用免疫组化法检测癌和癌旁组织中GOLPH3蛋白表达。采用Pearson线性相关分析miR-130a、GOLPH3 mRNA表达的相关性,统计分析miR-130a、GOLPH3 mRNA表达与患者临床病理参数的关系。采用Kaplan-Meier生存分析miR-130a、GOLPH3表达对NSCLC患者预后的影响,采用Cox回归分析影响NSCLC患者生存预后的危险因素。结果 与癌旁组织相比,NSCLC癌组织中miR-130a表达较低,而GOLPH3 mRNA及蛋白表达较高(P均<0.05)。NSCLC癌组织中miR-130a表达与GOLPH3 mRNA表达呈负相关(r=-0.457,P<0.05)。miR-130a、GOLPH3 mRNA表达与肿瘤TNM分期及淋巴结转移有关(P均<0.05)。miR-130a低表达、GOLPH3高表达的NSCLC患者3年生存率低于miR-130a高表达、GOLPH3低表达患者(P均<0.05)。TNM分期、淋巴结转移、miR-130a低表达及GOLPH3高表达是NSCLC患者不良预后的独立危险因素(P均<0.05)。结论 NSCLC癌组织中miR-130a表达降低、GOLPH3表达升高,检测二者表达有助于NSCLC病情判断及预后评估。
【Abstract】 Objective To study the expression and clinical significance of microRNA(miR)-130a and Golgi phosphoprotein-3(GOLPH3) in non-small-cell lung cancer(NSCLC) cancer tissues.Methods Eighty-seven NSCLC patients were selected as the research objects.Fluorescence quantitative PCR was used to detect the expression of miR-130 a and GOLPH3 mRNA in the cancer and adjacent tissues.Immunohistochemistry was used to detect the expression of GOLPH3 protein in cancer and adjacent tissues.Pearson linear correlation analysis was used to analyze the correlation between miR-130a and GOLPH3 mRNA expression.The relationship between miR-130a,GOLPH3 mRNA expression and clinicopathological parameters were analyzed statistically.Kaplan-Meier survival analysis was used to analyze the effects of miR-130 a,GOLPH3 expression on the prognosis of NSCLC patients.Univariate and multivariate Cox analysis was used to analyze the risk factors affecting the survival and prognosis of NSCLC patients.Results Compared with the adjacent tissues,the expression of miR-130a in NSCLC cancer tissues was significantly lower,while the expression levels of GOLPH3 mRNA and protein were significantly higher(all P<0.05).The expression of miR-130 a in NSCLC cancer tissues was negatively correlated with the expression of GOLPH3 mRNA(r=-0.457,P<0.05).The expression of miR-130 a and GOLPH3 mRNA was related to tumor TNM staging and lymph node metastasis(all P<0.05).The 3-year survival rate of NSCLC patients with low miR-130a expression and high GOLPH3 expression was significantly lower than that of patients with high miR-130 a expression and low GOLPH3 expression,respectively.TNM stage Ⅲ,lymph node metastasis,low expression of miR-130a and high expression of GOLPH3 were independent risk factors for the poor prognosis of NSCLC patients(all P<0.05).Conclusion The expression of miR-130a decreases and the expression of GOLPH3 increases in NSCLC cancer tissues;detection of them is helpful for the determination and prognosis evaluation of NSCLC.
