胃癌组织中miR-200a、miR-92a表达变化与患者临床病理参数和预后的关系
Expression changes of miR-200a and miR-92a in gastric cancer and their relationships with the clinicopathological parameters and prognosis of patients
【摘要】 目的 探讨胃癌组织中微小RNA(miR)-200a、miR-92a表达变化及与患者临床病理参数和预后的关系。方法 选择104例胃癌患者,采用RT-qPCR法检测胃癌及癌旁正常胃组织中miR-200a、miR-92a表达。分析miR-200a、miR-92a表达与胃癌患者临床病理参数的关系。Kaplan-Meier生存分析不同miR-200a、miR-92a表达胃癌患者生存情况,Cox回归分析胃癌患者预后的影响因素。结果 胃癌组织中miR-92a表达高于癌旁组织(P<0.05),miR-200a表达低于癌旁组织(P<0.05)。浸润至肌层、浆膜下及其外、T3-4a期、N1-3期胃癌患者癌组织中miR-200a表达低于浸润至黏膜及黏膜下层、T1-2期、N0期胃癌患者(P均<0.05),miR-92a表达高于浸润黏膜及黏膜下层、T1-2期、N0胃癌患者(P均<0.05)。miR-200a<1.49、miR-92a≥2.93胃癌患者3年总生存率、3年无瘤生存率低于miR-200a≥1.49、miR-92a<2.93胃癌患者(P均<0.05)。淋巴结转移、miR-200a<1.49、miR-92a≥2.93是胃癌患者预后不良的危险因素(P均<0.05)。结论 胃癌组织中miR-200a表达下调,miR-92a表达上调;检测二者表达有助于胃癌病情判断及预后评估。
【Abstract】 Objective To investigate the expression changes of microRNA-200a(miR-200a) and miR-92a in the gastric cancer and their relationships with the clinicopathological parameters and prognosis of patients.Methods Totally104 patients with gastric cancer were selected.The expression of miR-200a and miR-92a in the gastric cancer and adjacent normal gastric tissues were detected by RT-qPCR.We analyzed the relationship between the expression of miR-200a,miR-92a and clinicopathological parameters of gastric cancer.Kaplan Meier survival analysis was used to analyze the survival of gastric cancer patients with different miR-200a and miR-92a expression.Cox regression analysis was used to analyze the prognostic factors of gastric cancer patients.Results The expression of miR-92a in gastric cancer tissues was higher than that in the adjacent tissues(P<0.05),and the expression of miR-200a was lower than that in the adjacent tissues(P<0.05).The expression of miR-200a in gastric cancer tissues of patients with infiltration into muscle layer,sub serosal layer and outside,T3-4 a stage and N1-3 stage was lower than that in patients with infiltration into mucosa and submucosa,T1-2 stage and N0 stage(P<0.05),and the expression of miR-92a was higher than that in gastric cancer tissues of patients with infiltration into mucosa and submucosa,T1-2 stage and N0 stage(P<0.05).The 3-year overall survival rate and 3-year diseasefree survival rate of gastric cancer patients with miR-200a<1.49 and miR-92a≥2.93 were lower than those of patients with miR-200a≥1.49 and miR-92a<2.93(both P<0.05).Lymph node metastasis,miR-200a<1.49 and miR-92a≥2.93 were risk factors for poor prognosis of gastric cancer patients(all P<0.05).Conclusion MiR-200a expression is downregulated,miR-92a is up-regulated in the gastric cancer tissues,and detection of the expression of the two is helpful for the diagnosis and prognosis evaluation of gastric cancer.
