加味当归芍药散治疗后慢性子宫内膜炎大鼠子宫内膜胞饮突发育及NF-κb、TGF-β1蛋白表达变化
Changes in endometrial pinopodes and NF-κb and TGF-β1 protein expression in rats with chronic endometritis after treatment of modified Danggui Shaoyao powder
【摘要】 目的 观察加味当归芍药散灌胃治疗后慢性子宫内膜炎(CE)大鼠子宫内膜胞饮突发育情况及NF-κB、TGF-β1蛋白表达变化。方法 雌性SD大鼠60只,随机分为空白组、模型组、妇科千金胶囊组、中药高剂量组、中药中剂量组、中药低剂量组,每组10只。模型组、中药高剂量组、中药中剂量组、中药低剂量组、妇科千金胶囊组使用25%苯酚胶浆制备CE模型。中药高、中、低剂量组分别给予25.92、12.96、6.48 g/kg的加味当归芍药散灌胃;妇科千金胶囊组给予0.216 g/kg的妇科千金胶囊灌胃;空白组、模型组给予等量生理盐水灌胃。治疗21 d后,采集各组大鼠子宫组织,电镜观察子宫内膜胞饮突发育情况,Western blotting法检测NF-κB、TGF-β1蛋白。结果 与空白组相比,模型组大鼠子宫内膜表面胞饮突发育不成熟且数量少;与模型组相比,中药高、中剂量组大鼠子宫内膜表面胞饮突发育趋向成熟且数量较多,妇科千金胶囊组、中药低剂量组子宫内膜表面胞饮突发育程度中等且数量较少。模型组大鼠子宫组织中NF-κB、TGF-β1蛋白表达高于空白组,中药高、中剂量组和妇科千金胶囊组NF-κB、TGF-β1蛋白表达低于模型组(P均<0.01)。结论 加味当归芍药散可以改善CE大鼠子宫内膜胞饮突的发育情况,并下调NF-κB、TGF-β1蛋白表达水平。
【Abstract】 Objective To observe the changes in endometrial pinopodes and nuclear factor-κb(NF-κB) and transforming growth factor β1(TGF-β1) protein expression in rats with chronic endometritis(CE) after intragastric administration of modified Danggui Shaoyao powder.Methods Sixty female SD rats were randomly divided into the blank group,model group,gynecological Qianjin capsule group,herbal high-dose group,herbal medium-dose group and herbal low-dose group,with 10 rats in each group.The CE model was prepared by using 25% phenol mucilage in the model group,herbal high-dose group,herbal medium-dose group,herbal low-dose group and gynecological Qianjin capsule group.The rats in the herbal high-dose,medium-dose and low-dose groups were respectively given 25.92,12.96,and 6.48 g/kg modified Danggui Shaoyao powder;the rats in the gynecological Qianjin capsule group were given 0.216 g/kg gynecological Qianjin capsule;the rats in the blank group and model group were given equivalent normal saline.After 21 d of treatment,uterine tissues were collected from each group,and the endometrial pinopodes were observed by electron microscopy,and NF-κB and TGF-β1 proteins were detected by Western blotting.Results Compared with the blank group,the endometrial pinopodes in the model group were immature and less in number;compared with the model group,the endometrial pinopodes in the herbal high-dose group and herbal medium-dose group tended to be mature and more in number,while the endometrial pinopodes in the gynecological Qianjin capsule group and herbal low-dose group were moderately developed and less in number.The expression levels of NF-κB and TGF-β1 protein in the uterine tissues of rats in the model group were higher than those in the blank group;compared with the model group,the expression levels of NF-κB and TGF-β1 protein in the herbal high-dose group and herbal medium-dose group as well as the gynecological Qianjin capsule group were lower(all P<0.01).Conclusion The results shown that endometrial pinopodes can be improved and NF-κB and TGF-β1 protein expression levels can be down-regulated by the modified Danggui Shaoyao powder in rats with CE.
