枳实-白芍对C-IBS大鼠血清胃肠神经递质的影响
Effects of Zhishi(Aurantii Fructus Immaturus)-Baishao(Paeoniae Radix Alba) on Serum Gastrointestinal Neurotransmitters in Model Rats with C-IBS
【摘要】 目的:观察不同配伍比例的枳实 - 白芍对便秘型肠易激综合征(C-IBS)大鼠血清胃肠神经递质水平的影响,探讨其治疗 C-IBS 的可能机制。 方法:取 SPF 级 Wistar 成年雄性大鼠 60 只,随机留取 10 只大鼠作为空白组。 其余 50 只大鼠为实验组,均采用冰生理盐水灌胃的方法制备 C-IBS 模型,实验过程中 5 只大鼠脱落。 造模成功后将实验组大鼠随机分为模型对照组、匹维溴铵组(13.5 mg·kg-1)、枳实 - 白芍 1∶1 组(枳实0.82 g·kg-1,白芍 0.82 g·kg-1)、枳实 - 白芍 1∶2 组(枳实 0.82 g·kg-1,白芍 1.64 g·kg-1)、枳实 - 白芍 2∶1 组(枳实1.64 g·kg-1,白芍 0.82 g·kg-1),每组 9 只。 各组大鼠每天灌胃相应药物,空白组和模型对照组给予等量蒸馏水灌胃,持续 14 d。 给药结束后测定各组大鼠粪便相关指标以及血清中 5- 羟色胺(5-HT)、P 物质(SP)、降钙素基因相关肽(CGRP)、生长抑素(SS)、神经肽 Y(NPY)、内皮素(ET)的水平。 结果:与空白组比较,模型对照组大鼠的粪便性状评分、粪点数降低(P< 0.05),血清中 5-HT、SP、CGRP、SS 水平升高(P< 0.01),血清中 NPY、ET 水平降低(P< 0.01)。 与模型对照组比较,枳实 - 白芍不同配伍比例组粪便性状评分、粪点数均升高(P< 0.01 或P< 0.05),血清中 5-HT、SP、CGRP、SS 水平均降低(P< 0.01 或 P< 0.05),血清中 NPY、ET 水平均升高(P< 0.01)。结论:枳实 - 白芍治疗 C-IBS 的机制可能与降低血清中 5-HT、SP、CGRP、SS 的水平以及升高血清中 NPY、ET的水平,从而改善胃肠动力有关。
【Abstract】 Objective:To observe the effects of different compatibility proportion of Zhishi(Aurantii Fructus Immaturus)-Baishao(Paeoniae Radix Alba) on the serum levels of gastrointestinal neurotransmitters in rats with constipation irritable bowel syndrome(C-IBS),and to analyze its possible mechanism in the treatment of C-IBS. Methods:Total 60 adult male SPF Wistar rats were selected,and 10 rats were randomly selected as blank group. The other 50 rats served as the experimental group,and the C-IBS model was established by ice normal saline gavage method. During the experiment,5 rats fell off. After the model was prepared successfully,the rats in the experimental group were randomly divided into model control group,pinaverium bromide group(13.5 mg·kg-1),Zhishi-Baishao 1∶1 group(Zhishi:0.82 g·kg-1,Baishao:0.82 g·kg-1),Zhishi-Baishao 1∶2 group(Zhishi:0.82 g·kg-1,Baishao:1.64 g·kg-1) and Zhishi-Baishao 2∶1 group(Zhishi:1.64 g·kg-1,Baishao:0.82 g·kg-1),with 9 rats in each group. They were given the same drug by gavage every day. The blank and model control group were given equivalent distilled water by gavage for 14 days. After medication,feces related indexes and serum levels of5-hydroxytryptamine(5-HT),substance P(SP),calcitonin gene related peptide(CGRP),somatostatin(SS),neu ropeptide Y(NPY),endothelin(ET) were detected. Results:Compared with blank group,fecal trait scores and fecal count of rats in model control group were decreased(P< 0.05),and the serum levels of 5-HT,SP,CGRP and SS were increased(P< 0.01) and the serum levels of NPY and ET were decreased(P< 0.01).Compared with the model control group,different compatibility ratio Zhishi-Baishao groups all could increase fecal trait scores and fecal count(P< 0.01 or P< 0.05),and decrease the serum levels of 5-HT,SP,CGRP and SS(P< 0.01 or P< 0.05),and increase the serum levels of NPY and ET(P< 0.01). Conclusions:Zhishi-Baishao can treat C-IBS,its mechanism may be associated with decreasing the serum levels of 5-HT,SP,CGRP and SS,and increasing the serum levels of NPY and ET,so as to improve gastrointestinal motility.
