具有酪氨酸酶抑制作用的豌豆多肽酶解工艺研究
Study on enzymolysis process of pea polypeptide with tyrosinase inhibition
【摘要】 以豌豆分离蛋白为原料,采用酸性蛋白酶一次水解,水解条件为:加酶量5 000 U/g~6 000 U/g、酶解温度40℃~60℃,酶解时间90 min~120 min;采用NY50中性蛋白酶对酸性蛋白酶的水解液进行二次水解,对水解液进行膜分离,得到分子量300 Da~4 kDa的豌豆多肽,此多肽质量浓度在5 mg/mL~25 mg/mL范围时酪氨酸酶抑制率与多肽质量浓度呈量效关系。
【Abstract】 Pea protein isolate was used as raw material, and the hydrolysis conditions were as follows: adding enzyme amount was 5 000 U/g~6 000 U/g, enzymolysis temperature was 40 ℃~60 ℃, enzymolysis time was 90 min~120min; NY50 neutral protease was used to hydrolyze the hydrolysate of acid protease, and the hydrolysate was separated by membrane to obtain pea polypeptide with molecular weight of 300 DA~4 kDa. The tyrosinase inhibition rate of this polypeptide was dose-dependent on the polypeptide concentration in the range of 5 mg/L~25 mg/L.
