一株高效促生菌株的分离筛选与发酵条件初探
Isolation and Screening of a High-efficiency Growth-promoting Strain and Preliminary Exploration for Fermentation Conditions
【摘要】 为筛选能有效促进植物生长的促生细菌,从山东省泰安市泰山山脉常年生长植株根际土壤中分离获得一株芽孢杆菌TG-5。通过形态学特征和16S rRNA分析,将其鉴定为苏云金芽孢杆菌(Bacillus thuringiensis)。采用NBRIP培养基对溶磷能力进行测定,结果表明:苏云金芽孢杆菌TG-5对磷酸钙培养基溶磷效果较好,与对照相比,该菌株溶磷效率是其他菌株3倍以上,最高达10.7倍以上。通过对该菌株固体发酵条件初探,发现发酵培养基主要成分玉米粉和大豆粉的最佳配比为2:3、碳氮源分别为玉米粉和大豆粉时生物量最大。上述试验证明苏云金芽孢杆菌TG-5具有较好的促生应用潜力,可进一步扩大培养。
【Abstract】 In order to screen the growth-promoting bacteria that can effectively promote plant growth, a Bacillus TG-5 was isolated from the rhizosphere soil of perennial plants in Taishan Mountains, Tai’an City, Shandong Province. It was identified as Bacillus thuringiensis by morphological features and 16SrRNA analysis. The phosphorus-dissolving ability of NBRIP medium was measured. The results showed that B. thuringiensis TG-5 had better phosphorus-dissolving effect on calcium phosphate medium. 10.7 times or more. Through preliminary exploration of the solid fermentation conditions of this strain, it was found that the optimum ratio of the main components of the fermentation medium, corn meal and soybean meal, was 2:3,and the biomass was the largest when the carbon and nitrogen sources were corn meal and soybean meal, respectively. The above experiments proved that B. thuringiensis TG-5 has good growth-promoting application potential and can be further expanded.
