鲁中肉羊FecB基因多态性与体尺性状的关联分析

【摘要】 本研究旨在分析FecB基因g.746A>G位点多态性对鲁中肉羊生长性状的影响。以240只6月龄鲁中肉羊母羊为研究对象,通过一代测序技术和琼脂糖凝胶电泳对鲁中肉羊FecB基因该位点进行多态性检测,采用单因素方差分析不同基因型与鲁中肉羊生长性状的关系。结果表明,FecB基因g.746A>G位点存在3种基因型,BB、B+和++,且B+为优势等位基因型,B为优势等位基因。遗传参数分析结果表明,FecB基因g.746A>G位点的杂合度是0.498,纯和度是0.502,有效等位基因数是1.992,多态信息含量是0.374,表现为中度多态性(0.25<PIC<0.5)。关联分析结果表明,FecB基因g.746A>G位点BB基因型的体高显著低于B+和++基因型(P<0.05);BB基因型的胸围和腹围显著低于++基因型(P<0.05),极显著低于B+基因型(P<0.01);该位点三种基因型的体长、管围、尻宽、尻长、胸深、胸宽、腰角宽、坐骨端宽均无显著差异(P>0.05)。综上所述,FecB基因突变位点对鲁中肉羊生长性状有一定程度的影响,本研究可为FecB基因在鲁中肉羊的分子标记辅助育种提供理论参考依据。

【Abstract】 The purpose of this study was to analysis the effect of the polymorphism of g.746a > G locus in FecB gene on the growth traits of Luzhong mutton sheep. A total of 240 6-month-old Luzhong mutton sheep were selected, the polymorphism of FecB gene was detected using sanger sequencing technology and agarose gel electrophoresis, and the correlation between different genotypes and growth traits was analyzed by one-way ANOVA. The results showed that there were three genotypes at g.746a > G locus of FecB Gene, BB, B+ and ++, the B+ was the dominant genotype, and B was the dominant allele. The results of genetic parameter analysis indicated that the heterozygosity of g.746a > G locus was 0.498, the homozygosity was0.502, the number of effective alleles was 1.992, and the content of polymorphism information was 0.374, showing a moderate polymorphism(0.25< PIC <0.5). The results of association analysis showed that the body height of BB genotype at g.746a > G locus was significantly lower than that of B+ and ++(P<0.05); The chest circumference and abdominal circumference of BB genotype were significantly lower than those of ++ genotype(P<0.05), and very significantly lower than those of B+ genotype(P<0.01); There was no significant difference in body length, tube circumference, rump width,rump length, chest depth, chest width, waist angle width and sciatic end width between the three genotypes(P >0.05). In conclusion, the mutationof FecB Gene have a certain impact on the growth traits of Luzhong mutton sheep, which provides a reference basis for molecular marker assisted breeding of FecB Gene in Luzhong mutton sheep.
