藏药八味沉香散血清对H9c2心肌细胞缺氧/复氧损伤的保护作用
Protective effects of serum of Tibetan medicine Bawei Chenxiang Powder on the hypoxia/reoxygenation injury of H9c2 myocardial cells
【摘要】 目的 研究藏药八味沉香散血清对H9c2心肌细胞缺氧/复氧损伤的保护作用。方法 将H9c2心肌细胞置于三气培养箱缺氧处理8 h,然后更换新鲜高糖培养基做常规培养(4h),建立缺氧/复氧模型;将实验组分为空白组,缺氧/复氧模型组,八味沉香散血清低、中、高剂量组及复方丹参片(阳性对照)组,空白组及模型组加入空白血清,含药组加入含药血清,进行缺氧/复氧干预,待干预结束后,在倒置显微镜下观察细胞生长及形态变化;用CCK-8法检测细胞存活率;用微板法和ELISA法分别检测缺氧8 h和复氧4 h后细胞培养上清液中乳酸脱氢酶(LDH)和磷酸肌酸激酶(CK)活性。结果 与模型组相比,含药血清低、中、高剂量组镜下心肌细胞形态相对完整;与模型组相比,含药血清低、中、高剂量组细胞存活率均显著升高(P<0.05)。与空白组相比,缺氧8 h及复氧4 h后模型组上清液中LDH、CK的活性显著升高(P<0.05);与模型组相比,含药血清中剂量组LDH显著降低(P<0.05),含药血清中、高剂量组CK显著降低(P<0.05)。结论 藏药八味沉香散血清对H9c2心肌细胞缺氧/复氧损伤具有一定的保护作用。
【Abstract】 Objective To study the protective effects of serum of Tibetan medicine Bawei Chenxiang Powder on the hypoxia/reoxygenation injury of H9 c2 myocardial cells.Methods H9 c2 myocardial cells were placed in a three-gas incubator for hypoxia treatment for 8 h, and then replaced with fresh high-glucose medium for routine culture for 4 h to establish the hypoxia/reoxygenation model; the experiment was divided into blank group, hypoxic/reoxygenated model group, low-dose, medium-dose and high-dose groups of Bawei Chenxiang Powder serum and compound Danshen Tablet(positive control)group.The blank group and model group were added with blank serum.The drug-containing group was added with drug containing serum for hypoxia/reoxygenation intervention.The cell growth and morphological changes were observed under an inverted microscope after the intervention; the cell survival rate was detected by CCK-8 method; The activities of lactate dehydrogenase(LDH)and phosphocreatine kinase(CK)in supernatant of cell culture after 8 h hypoxia and 4 h reoxygenation were determined by microplate method and elisa method, respectively.Results The morphology of myocardial cells in low-dose, medium-dose and high-dose groups of Bawei Chenxiang Powder serum was relatively intact compared with model group; the cell survival rate in low-dose, medium-dose and high-dose groups of Bawei Chenxiang Powder serum was significantly increased compared with model group(P<0.05);compared with blank group, the activities of LDH and CK in the supernatant of model group were increased significantly(P<0.05)after 8 hours of hypoxia and 4 hours of reoxygenation; LDH in medium-dose group of Bawei Chenxiang Powder serum was significantly decreased(P<0.05)compared with model group; CK in medium-dose and high-dose groups of Bawei Chenxiang Powder serum were significantly decreased(P<0.05).Conclusion The serum of Tibetan medicine Bawei Chenxiang Powder can protect H9 c2 myocardial cells from hypoxia/reoxygenation injury.
