实时荧光定量PCR技术在婴儿HIV感染早期诊断中的应用
Application of real-time PCR assay for early infant diagnosis of HIV infection in Yunnan
【摘要】 目的 评价实时荧光定量PCR技术作为婴儿HIV感染早期诊断方法的可行性。方法 2019年4月至2020年8月对大理州和德宏州出生的HIV暴露婴儿51例在五个阶段采集干血斑,用实时荧光定量PCR检测HIV-1DNA。对6周龄干血斑同时进行罗氏HIV-1DNA定性检测。同期对21例已知HIV感染婴儿的干血斑进行了检测。结果 共对送检的51例婴儿的235份干血斑进行了检测,结果全部是HIV-1DNA阴性,与罗氏HIV-1核酸定性结果一致。21例已知感染婴儿的干血斑实时荧光定量PCR检测结果全部是HIV-1DNA阳性(病毒载量均值为4428 copies/1x106cells)。结论 实时荧光定量PCR技术可以准确诊断暴露婴儿的HIV感染情况,为及时实施抗病毒治疗提供科学依据。
【Abstract】 Objective To evaluate the real-time PCR assay as a method in early infant diagnosis(EID)of HIV infection. Methods Dried blood spots(DBS) were collected from HIV exposed infants at five stages in Dali and Dehong prefectures and sent to test HIV-1DNA with the real-time PCR assay in Yunnan Center for Disease Control and Prevention from April 2019 to August 2020. The DBS from six month-old infants were also tested with Roche HIV-1 test kit. In the mean time,21 stored DBS of HIV infected infants were tested. Results 235 DBS collected from 51 infants were tested and the results were all HIV-1 DNA negative,which was consistent with Roche kit. 21 stored DBS of HIV infected infants were all HIV-1 DNA positive with the mean viral loads of 4428 copies/1x106cells. Conclusion The real-time PCR assay can diagnosis exposed infants correctly and provide scientific evidence for instant treatment and care for infected infants.
