白细胞介素-11促进巨噬细胞M2极化及对痛风性关节炎的保护作用
Interleukin-11 promotes macrophage M2 polarization and alleviates acute gouty arthritis in mice
【摘要】 目的 探讨白细胞介素(IL)-11在痛风性关节炎(GA)中的作用及其机制。方法 45只C57BL/6雄性小鼠,随机分为对照组、模型组和IL-11组。微晶尿酸钠注射用于构建小鼠GA模型,造模后IL-11组小鼠腹腔注射IL-11重组蛋白(20μg/kg)。7 d后进行踝关节肿胀度和功能障碍测定,评估踝关节病理形态;检测血清UA、XO、NO、IL-1β、IL-4、IL-6和IL-10含量;检测小鼠滑膜组织巨噬细胞极化和iNOS、IL-1β、Arginase-1和IL-10 mRNA表达。结果 HE结果显示,模型组小鼠滑膜增生明显,大量炎性细胞浸润;IL-11组小鼠滑膜增生和炎性细胞较模型组显著减少;与模型组比较,IL-11组小鼠炎症评分、踝关节肿胀度和功能障碍评分均显著降低(P<0.05);与模型组比较,IL-11组小鼠UA和XO含量显著降低,NO含量升高(P<0.05);与模型组相比,IL-11组小鼠M2/M1比值、Arginase-1和IL-10 mRNA显著升高,iNOS和IL-1β mRNA表达降低(P<0.05);与模型组相比,IL-11组小鼠血清IL-1β和IL-6表达明显降低,IL-4和IL-10表达明显升高(P<0.05)。结论 IL-11通过调控巨噬细胞极化,抑制关节炎症,有效缓解GA病程。
【Abstract】 To investigate the role of interleukin-11(IL-11) in gouty arthritis(GA), total of 45 male mice were recruited and randomly divided into control group, model group and IL-11 group. GA model was constructed by using monosodium urate(MUS). Mice in each group were treated with corresponding reagent continuously for 7 days,then the ankle joint swelling and dysfunction were measured, and serum UA, XO, NO, IL-1β, IL-4, IL-6 and IL-10levels and the mRNA expression of iNOS, IL-1β, Arginase-1 and IL-10 were evaluated. Compared with the control group, the inflammation score, ankle joint swelling, dysfunction scores, and the contents of UA and XO in the model group were significantly increased, while the contents of NO were decreased(P<0.05); compared with the model group, the indexes mentioned above were reversed in the IL-11 group(P<0.05). Compared with the control group,the M2/M1 ratio, Arginase-1 and IL-10 mRNA, and the expression of IL-4 and IL-10 in the model group were significantly reduced, while the expression of iNOS and IL-1β mRNA, and the expression of serum IL-1β and IL-6was increased(P<0.05); compared with the model group, the indexes mentioned above were reversed in the IL-11group(P<0.05). In conclusion, IL-11 could effectively alleviate gouty arthritis by regulating the polarization of macrophages and inhibiting the expression of inflammatory factors.
