miR-30c-2过表达联合阿霉素对甲状腺乳头状癌细胞增殖及迁移能力的影响
Effects of mi R-30c-2 overexpression combined with adriamycin on the proliferation and migration of thyroid papillary carcinoma cells
【摘要】 目的 探究微小RNA-30c-2(microRNA-30c-2,miR-30c-2)联合阿霉素对甲状腺乳头状癌TPC-1细胞增殖和迁移能力的影响。方法 体外培养TPC-1细胞,将细胞分为空白对照组(未做任何处理的细胞,即Control组)、过表达对照组(瞬时转染NC mimic质粒,即NC mimic组)、过表达组(瞬时转染miR-30c-2过表达质粒,即miR-30c-2组),采用qRT-PCR技术检测细胞中miR-30c-2的表达水平;将TPC-1细胞分为4组,分别为空白对照组(未做任何处理的细胞,Control组)、阿霉素处理组(使用阿霉素混合液培养TPC-1细胞24 h、48 h)、过表达组(瞬时转染miR-30c-2过表达质粒,即miR-30c-2 mimic组)、共处理组(阿霉素混合液与转染miR-30c-2过表达质粒共同培养TPC-1细胞24 h、48 h),CCK-8试验及细胞划痕实验分别检测4组细胞的增殖和迁移能力。结果 经qRT-PCR检测,与NC mimic组相比,过表达组的miR-30c-2表达水平明显升高(P<0.001);CCK-8实验表明,阿霉素+miR-30c-2 mimic共处理组分别与阿霉素处理组和过表达组相比,可以明显抑制细胞增殖(P<0.05);细胞划痕实验表明,阿霉素+miR-30c-2 mimic共处理组对比阿霉素组和过表达组发现TPC-1细胞迁移能力进一步受到抑制(P<0.05)。结论 miR-30c-2过表达可以联合阿霉素共同抑制甲状腺乳头状癌TPC-1细胞的增殖和迁移能力。
【Abstract】 Objective To investigate the effects of microRNA-30 c-2(miR-30 c-2) combined with adriamycin on the proliferation and migration of thyroid papillary carcinoma TPC-1 cells. Methods TPC-1 cells were cultured in vitro. The cells were divided into blank control group(cells without any treatment, i.e. control group), overexpression control group(transient transfection of NC mimic plasmid, i.e. NC mimic group) and overexpression group(transient transfection of miR-30 c-2 overexpression plasmid, i.e. miR-30 c-2 group). The expression level of miR-30 c-2 in the cells was detected by qRT-PCR. TPC-1 cells were divided into four groups: blank control group(cells without any treatment, control group), doxorubicin treatment group(TPC-1 cells were cultured with doxorubicin mixture for 24 h and 48 h), overexpression group(transient transfection of miR-30 c-2 overexpression plasmid, i.e. miR-30 c-2 mic group), CO treatment group(TPC-1 cells were cultured with doxorubicin mixture and transient transfection of miR-30 c-2 overexpression plasmid for 24 h and 48 h), CCK-8 test and cell scratch test were used to detect the proliferation and migration of the four groups of cells.Results qRT-PCR showed that the expression level of miR-30 c-2 in overexpression group was significantly higher than that in NC mic group(P<0.001). CCK-8 experiment showed that adriamycin + miR-30 c-2 mic CO treatment group could significantly inhibit cell proliferation compared with adriamycin treatment group and overexpression group, respectively(P<0.05). Cell scratch test showed that the migration ability of TPC-1 cells was further inhibited in adriamycin+miR-30 c-2 mic CO treatment group compared with adriamycin group and overexpression group(P<0.05). Conclusion Overexpression of miR-30 c-2 combined with adriamycin can inhibit the proliferation and migration of thyroid papillary carcinoma TPC-1 cells.
