干扰LncRNA OIP5-AS1对人乳腺癌细胞迁移、上皮间充质转化及Notch1表达的影响
Effects of interfering LncRNA OIP5-AS1 on breast cancer cell migration, epithelial mesenchymal transformation and Notch1 expression
【摘要】 目的 探讨干扰LncRNAOIP5-AS1对人乳腺癌细胞MDA-MB-231迁移、上皮间充质转化及Notch1表达的影响。方法 培养MDA-MB-231细胞,转染LncRNA OIP5-AS1 siRNA为LncRNA OIP5-AS1 siRNA组,转染NC siRNA的为NC siRNA组,未作任何处理的为Control组;采用RT-qPCR方法检测各组细胞LncRNAOIP5-AS1的表达水平;采用细胞划痕实验检测各组细胞迁移能力;Western blots法检测LncRNA OIP5-AS1被干扰后乳腺癌细胞上皮间充质转化相关蛋白E-cadherin和Vimentin的表达变化;T-qPCR方法检测Notch1 mRNA的表达水平。结果 通过siRNA技术使LncRNA OIP5-AS1的表达受到抑制;LncRNA OIP5-AS1被干扰后,乳腺癌细胞的迁移能力降低,与Control组和NC siRNA组比,LncRNA OIP5-AS1 siRNA组细胞迁移能力受抑制更加明显(P<0.01);Western blots结果显示:与Control组和NC siRNA组相比,LncRNA OIP5-AS1 siRNA组细胞的E-cadherin蛋白表达水平升高(P<0.05),而Vimentin蛋白表达水平降低(P<0.05);Notch1 mRNA的表达也受到抑制(P<0.01)。结论 LncRNA OIP5-AS1很可能通过对Notch信号通路的调控,从而实现对乳腺癌细胞的迁移能力及上皮间充质转化的抑制作用。
【Abstract】 Objective To investigate the effects of interfering LncRNA OIP5-AS1 on migration, epithelial mesenchymal transformation and Notch1 expression of MDA-MB-231 breast cancer cells. Methods LncRNA OIP5-AS1 in breast cancer cells was silenced by small interfering RNA(siRNA) technology. The experiment was divided into three groups: Control group, negative control(NC) siRNA group and LncRNAOIP5-AS1 siRNA group. RT-qPCR was used to detect the interfered effects of LncRNAOIP5-AS1 in all groups. Cell migration was detected by scratch test. After LncRNA OIP5-AS1 was downregulated, Western blots was used to detect the expression of E-cadherin and Vimentin protein in MDA-MB-231 breast cancer cells, and further, the expression of Notch1 mRNA was detected by RT-qPCR.Results LncRNA OIP5-AS1 was successfully interfered in LncRNAOIP5-AS1 siRNA group. After LncRNA OIP5-AS1 was interfered, the migration ability of MDA-MB-231 breast cancer cells was decreased, which was more significantly inhibited in the interference group than that in the Control group and NC siRNA group(P<0.01). In LncRNA OIP5-AS1 siRNA group, the expression of E-cadherin protein was increased while the Vimentin protein was decreased. Further experiments showed that the Notch1 mRNA was also inhibited, and the Notch1 mRNA decreased more significantly in the interference group than that in the Control group and the NC siRNA group(P<0.01).Conclusion Interference of LncRNA OIP5-AS1 can significantly inhibit the migration and the epithelial mesenchymal transformation of MDA-MB-231 breast cancer cells and inhibit the expression of Notch1 mRNA, which also reflects that LncRNA OIP5-AS1 probably regulates notch signaling pathway to inhibit the above effects.
