响应面法优化豌豆皮中的总酚提取工艺研究

【摘要】 采取溶剂浸提法提取豌豆皮的总酚。以乙醇为提取剂,以料液比、乙醇体积分数、浸提时间、浸提温度为影响因素,以总酚提取量为响应值,在单因素试验的基础上,利用响应面法优化豌豆皮总酚的提取工艺,并通过测定DPPH·、ABTS~+·清除率反映其体外抗氧化活性。结果表明:豌豆皮总酚的最佳提取条件为料液比1∶60 (g/mL)、乙醇体积分数50%、浸提时间3.7 h、浸提温度70℃,在此条件下,豌豆皮总酚提取量为3.981 5 mg/g, DPPH·清除率为88.56%,ABTS~+·清除率为74.75%。

【Abstract】 The total phenol of pea peel was extracted by solvent extraction method. Ethanol was used as the extraction agent, and the material liquid ratio, ethanol volume fraction, extraction time and extraction temperature were used as the influencing factors. On the basis of single factor test, the extraction process of total phenols of pea peel was optimized by response surface method, and its antioxidant activity in vitro was reflected by measuring the scavenging rate of DPPH·, ABTS~+·. The results showed that the best extraction conditions of total phenols from pea peel were as follows: solid-liquid ratio 1 ∶60(g/mL), ethanol volume fraction 50 %, extraction time 3.7 h, extraction temperature 70 ℃. Under these conditions, the extraction amount of total phenols from pea peel was 3.981 5 mg/g, and the scavenging rate of DPPH· and ABTS~+· was 88.56 % and 74.75 %, respectively.
