新疆边境昆玉市亚洲璃眼蜱形态及分子生物学鉴定
Morphological classification and molecular identification of Hyalomma asiaticum in Kunyu of Xinjiang
【摘要】 试验旨在了解新疆边境地区昆玉市的蜱种分布状况。从该地区骆驼体表采集寄生蜱虫,应用数码体视显微镜初步进行形态学鉴定,采用蜱种鉴定的特异性引物对总DNA进行分子生物学检测,采用MEGA 10.0软件邻接法构建遗传进化树,确定蜱虫的进化生物学特征。结果表明,从昆玉市骆驼体表采集的蜱样符合亚洲璃眼蜱的形态学特征,线粒体16S rDNA PCR产物的测序结果表明,采集蜱虫为亚洲璃眼蜱;采集蜱在系统发育上与新疆南疆地区尉犁县采集的亚洲璃眼蜱亲缘关系较近,而与北疆石河子、福海以及疆外内蒙古鉴定株的亲缘关系较远。研究表明,昆玉市骆驼体表寄生的亚洲璃眼蜱初步呈现地域差异的进化规律性。
【Abstract】 The experiment aims to understand the distribution of tick species in Kunyu City, Xinjiang border region.Collecting parasitic ticks from camels in the area, preliminary morphological identification was carried out by digital stereo microscope, molecular biology detection of total DNA was carried out using specific primers for identification of tick species, and genetic evolutionary tree was constructed by MEGA 10.0 software neighbor-joining method to determine the evolutionary biological characteristics of ticks. The results showed that the tick samples collected from the body surface of camels in Kunyu City conformed to the morphological characteristics of Asian glass eyes, and the sequencing results of mitochondrial 16 S rDNA PCR products showed that the collected ticks were Asian glass eyes. The phylogeny of the collected ticks was relatively close to the Asian glass eye tick collected from Yuli County in southern Xinjiang, but far from Shihezi and Fuhai in northern Xinjiang and the identified strains in Inner Mongolia outside Xinjiang. The study indicates that the Asian tick parasitic on camels in Kunyu City initially shows the evolutionary regularity of regional differences.
