【摘要】 目的 探讨长链非编码RNA NEAT1与微小RNA-128在老年高血压患者血清中的表达及其临床意义。方法 将86例高血压患者设为研究组，根据高血压分级标准分为高血压1级(32例)、高血压2级(29例)、高血压3级(25例),选取46名健康老年人设为对照组。检测两组血清长链非编码RNA NEAT1、微小RNA-128表达，测量收缩压、舒张压，检测血清同型半胱氨酸、胱蛋白酶抑制剂C、视黄醇结合蛋白、β2微球蛋白、尿素、肌酐水平，分析老年高血压患者血清长链非编码RNA NEAT1、微小RNA-128表达与血压、同型半胱氨酸、胱蛋白酶抑制剂C、肾功能指标的相关性。结果 研究组收缩压、舒张压及血清同型半胱氨酸、胱蛋白酶抑制剂C、视黄醇结合蛋白、β2微球蛋白、尿素、肌酐水平均显著高于对照组(P<0.05);高血压2级患者收缩压、舒张压及血清同型半胱氨酸、胱蛋白酶抑制剂C、视黄醇结合蛋白、β2微球蛋白、尿素、肌酐水平均显著高于1级患者(P<0.05),3级患者显著高于1级、2级患者(P<0.05)。研究组长链非编码RNA NEAT1表达显著高于对照组(P<0.05),微小RNA-128表达显著低于对照组(P<0.05)。高血压2级患者长链非编码RNA NEAT1表达显著高于1级患者(P<0.05),3级患者显著高于1级、2级患者(P<0.05)。高血压2级患者微小RNA-128表达显著低于1级患者(P<0.05),3级患者显著低于1级、2级患者(P<0.05)。高血压患者长链非编码RNA NEAT1表达与收缩压、舒张压及血清同型半胱氨酸、胱蛋白酶抑制剂C、视黄醇结合蛋白、β2微球蛋白、尿素、肌酐水平呈显著正相关(P<0.05),微小RNA-128表达与收缩压、舒张压及血清同型半胱氨酸、胱蛋白酶抑制剂C、视黄醇结合蛋白、β2微球蛋白、尿素、肌酐水平呈显著负相关(P<0.05)。高血压患者血清长链非编码RNA NEAT1表达与微小RNA-128表达呈显著负相关(P<0.05)。结论 长链非编码RNA NEAT1在老年高血压患者血清中表达上调，微小RNA-128表达下调，二者与患者血压、肾功能受损密切相关，可能通过相互作用共同参与高血压及肾损害过程，并可能作为高血压患者肾功能受损的重要辅助指标。更多还原

【Abstract】 Objective To investigate the expression of long non-coding RNA NEAT1(LncRNA NEAT1) and microRNA-128(miR-128) in serum of elderly patients with hypertension and its clinical significance. Methods A total of 86 patients with hypertension were selected as the study group. According to the hypertension classification criteria, the elderly patients were divided into grade 1 hypertension(32 cases), grade 2 hypertension(29 cases), and grade 3 hypertension(25 cases). Forty-six healthy elderly people were selected as the control group. The expressions of LncRNA NEAT1 and miR-128 in serum of the two groups were detected, and the systolic blood pressure(SBP) and diastolic blood pressure(DBP) were measured.Serum levels of homocysteine(Hcy),cystatin C(Cyst C),retinol binding protein,β2 microglobulin,urea and creatinine were detected.The correlation between the expression of serum LncRNA NEAT1,miR-128 and blood pressure,Hcy,Cyst C,renal function indexes was analyzed in elderly patients with hypertension.Results Compared with the control group,the levels of SBP,DBP,Hcy,Cyst C,retinol binding protein,β2 microglobulin,urea and creatinine in the study group were significantly higher(P<0.05).The levels of SBP,DBP,Hcy,Cyst C,retinol binding protein,β2 microglobulin,urea and creatinine of patients with grade were significantly higher than those of patients with grade 1 hypertension(P<0.05),and those of patients with grade 3 hypertension were significantly higher than those of patients with grade 1 hypertension and grade (P<0.05).The expression of LncRNA NEAT1 in the study group was significantly higher than that in the control group(P<0.05),and the expression of miR-128 in the study group was significantly lower than that in the control group(P<0.05).The expression of LncRNA NEAT1 in patients with grade 2 hypertension was significantly higher than that in patients with grade 1 hypertension(P<0.05),and that in patients with grade 3 hypertension was significantly higher than that in patients with grade 1 hypertension and grade 2 hypertension(P<0.05).The expression of miR-128 in patients with grade 2 hypertension was significantly lower than that in patients with grade 1 hypertension(P<0.05),and that in patients with grade 3hypertension was significantly lower than that in patients with grade 1 hypertension and grade 2 hypertension(P<0.05).The expression of LncRNA NEAT1 was positively correlated with SBP,DBP,serum Hcy,Cyst C,retinol binding protein,β2 microglobulin,urea and creatinine in elderly patients with hypertension(P<0.05).The expression of miR-128 was negatively correlated with SBP,DBP,serum Hcy,Cyst C,retinol binding protein,β2 microglobulin,urea and creatinine(P<0.05).The expression of LncRNA NEAT1was negatively correlated with the expression of miR-128in elderly patients with hypertension(P<0.05).Conclusions The expression of LncRNA NEAT1 is up-regulated and miR-128 is down-regulated in the serum of elderly patients with hypertension,which is closely related to blood pressure and renal function impairment in patients,and may be involved in the process of hypertension and renal damage through interaction,and may serve as an important auxiliary indicator of renal function impairment in hypertensive patients.更多还原
