【摘要】 目的 初步评估应用液相色谱串联质谱技术(LC-MS/MS)对病原菌进行鉴定的可行性,为该技术临床应用提供实验依据。方法 以全基因基础上的蛋白质组学库作为通用细菌鉴定数据库,用LC-MS/MS技术对41株标准菌株以及临床分离的22株奈瑟菌和46株大肠埃希菌进行鉴定。结果 串联质谱技术鉴定病原菌在种的水平鉴定准确率为100%。以13株标准菌株样品稀释倍数和检测获得的肽段数量进行线性分析发现,二者之间具有很好的线性关系,相关系数均在0.90以上。结论 LC-MS/MS有望成为病原菌鉴定检测新方法。

【Abstract】 Objective To evaluate the feasibility of the identification for pathogens by tandem mass spectrometry(liquid chromatography-tandem mass spectrometry, LC-MS/MS) and to provides its experimental basis in clinical application. Methods A proteomic database based on the whole gene was established as a general database for bacterial identification, and 41 standard strains, 22 strains of Neisseria and 46 strains of Escherichia coli isolated from clinical samples were identified by the established LC-MS/MS. Results The accuracy of the identification for pathogens at species level by tandem mass spectrometry was 100%. Based on the linear analysis of the dilution ratio of the samples from 13 standard strains and the detectable number of peptides, a good linear relationship was found between the two methods with above 0.90 of correlation coefficients. Conclusion LC-MS/MS should be promising to become a new method for identification and detection of pathogens.
