【摘要】 目的:通过小鼠脓毒症模型的建立和评价,为研究脓毒症的发病机制和治疗措施提供可稳定重复的模型动物。方法:CLP法建立小鼠脓毒症模型;从术后小鼠体质量、生存状况,血常规、血生化,血液及腹腔灌洗液载菌量,血清IL-6和TNF-α水平,组织HE染色等指标对模型进行评价。结果:60%的小鼠术后72h内死亡,体质量明显下降(P<0.05);CLP术后,小鼠的血液和腹腔洗液载菌量呈阳性,血清IL-6、TNF-α水平显著上调(P<0.05),血常规、血生化明显改变,肝、肾、肺和脾等组织出现了明显病理损伤。结论:本研究成功建立CLP脓毒症模型。

【Abstract】 Objective:Through the establishment and evaluation of mouse sepsis model,to provide stable and repeatable model animals for studying the pathogenesis and treatment of sepsis.Methods:CLP method was used to establish a mouse sepsis animal model,and the model was evaluated from the postoperative mouse weight change,survival state,blood routine,blood biochemical test,blood and peritoneal fluid bacterial culture,serum IL-6 and TNF-αlevel and the histopathological changes by HE staining.Results:60% of the mice died within 72 hours after operation,and their body weight decreased significantly(P<0.05).Bacteria in blood and abdominal lotion were positive.Serum IL-6 and TNF-α levels were significantly up-regulated(P <0.05).Significant changes in blood routine and blood biochemistry;Liver,kidney,lung,spleen and other tissues showed obvious pathological damage.Conclusion:The sepsis model of CLP was successfully established in this study.
