二至丸改善D-gal致衰老小鼠学习记忆障碍的海马代谢组学研究
Hippocampus Metabolomic Study on Improving Learning and Memory Impairment of Erzhi Pill in D-gal-induced Aging Mice
【摘要】 目的 研究二至丸改善D-gal致衰老小鼠学习记忆障碍的作用机制。方法 采用D-gal诱导衰老复制阿尔茨海默病模型,实验分为空白组、模型组、二至丸组。Morris水迷宫实验观察各组小鼠学习记忆能力,HE染色观察海马神经元细胞形态,并通过GC-MS代谢组学技术结合多元统计方法对小鼠海马组织行分析,筛选内源性差异代谢产物。结果 与模型组相比,二至丸组小鼠学习记忆能力恢复正常水平,海马神经元形态改善;二至丸可上调海马组织中N-乙酰氨基葡萄糖、D-谷氨酰胺、L-酪氨酸、甘油-3-磷酸、DL-丝氨酸表达水平(P <0.05),下调L-半胱氨酸亚磺酸、N-乙酰丝氨酸表达水平(P <0.05)。结论 二至丸能够改善D-gal诱导衰老小鼠的学习记忆能力,改善海马神经元形态,通过改善大脑能量代谢障碍、抗氧化应激、调节神经突触可塑性等多方面实现神经保护作用。
【Abstract】 Objective To study the mechanism of Erzhi pill on improving learning and memory impairment in D-gal-induced aging mice by non-targeted metabolomics technology. Methods Alzheimer’s disease model was induced by D-gal. The experiment were divided into control group, model group and EZW group. Morris water maze test was used to observe the learning and memory ability of mice in each group, and HE staining was used to observe the morphology of hippocampal neurons. GC-MS metabolomics technology combined with multivariate statistical method was used to analyze the hippocampus of mice, and the endogenous differential metabolites were screened. Results Compared with the model group, the learning and memory ability of mice in the EZW group returned to normal level.Erzhi pill can up-regulate the expression levels of N-Acetyl-D-glucosamine, D-glutamine, L-tyrosine, glycerol-3-phosphate and DL-serine in hippocampus(P<0.05), down-regulate L-cysteine sulfinic acid and N-acetylserine expression levels(P<0.05). Conclusion Erzhi pill can improve the learning and memory ability of D-gal-induced aging mice and improve the morphology of hippocampal neurons. Its mechanism is to achieve neuroprotective effect by improving brain energy metabolism disorder, regulating synaptic plasticity, and resisting oxidative stress.
