石油烃降解菌LYC-1和LYC-2的筛选及其降解效能研究

【摘要】 从大庆某湖泊底泥中分离出两株能以石油烃类物质为唯一碳源的石油烃降解菌株,分别命名为LYC-1、LYC-2。经过微生物形态、生理生化特性及16S rDNA测序分析,确定LYC-1为莫氏不动杆菌(Acinetobacter modestus);LYC-2为铜绿假单胞菌(Pseudomonas aeruginosa)。研究了两株菌的降解特性,测定了两菌株在不同培养基中的生长情况,并利用单因素试验考察了多种环境条件因子对两菌株降解效能的影响。结果表明,菌株LYC-1在pH为8、温度25℃、溶解氧6.5 mg/L、接种量5%的条件时有利于其对石油烃类的降解。菌株LYC-2在pH为7、温度30℃、溶解氧5.0 mg/L、接种量5%等条件下有利于其对石油烃类的降解。

【Abstract】 Two petroleum hydrocarbon degradation strains, named LYC-1 and LYC-2, were isolated from the sediment of a lake in Daqing. Through the analysis of the morphology, physiological and biochemical characteristics of the microorganism and 16S rDNA sequencing, LYC-1 was determined to be Acinetobacter modestus. Lyc-2 is Pseudomonas aeruginosa. In this experiment, the degradation characteristics of the two strains were studied, the growth of the two strains in different media was measured, and the effects of various environmental factors on the degradation efficiency of the two strains were investigated by single factor test. The results showed that the pH of LYC-1 was 8, the temperature was 25 ℃, the dissolved oxygen was 6.5 mg/L, and the inoculum size was 5%, which was favorable for the degradation of petroleum hydrocarbons. Under the conditions of pH 7, temperature 30 ℃, dissolved oxygen 5.0 mg/L and inoculum size 5%, LYC-2 was beneficial to the degradation of petroleum hydrocarbons.
