15位酰腙类苦参碱衍生物的合成以及抗菌活性研究
Synthesis and antibacterial activity of matrine derivatives from 15 acylhydrazone
【摘要】 以苦参碱为原料,在三氯氧磷的作用下,设计合成了12个苦参碱腙类衍生物,产率在60~90%之间。所有化合物均通过1H-NMR、13C-NMR和质谱数据进行了表征。该合成方法操作简单,适用范围广,后处理容易。抗菌实验采用二倍稀释法检测15位苦参碱腙类衍生物分别对两种革兰氏阳性菌细菌、革兰氏阴性菌以及一种真菌的抑制活性,希望能找到抗菌活性较好的药物候选物。结果显示大部分目标化合物具有不同程度的抗菌活性,其中B4和B10的抗菌效果较好,对大肠杆菌、绿脓杆菌、金黄色葡萄球菌、痤疮丙酸杆菌和白色念珠菌MIC值分别为:B4:1mg·mL-1、1 mg·mL-1、0.5 mg·mL-1、0.25mg·mL-1、0.25mg·mL-1;B10:2 mg·mL-1、4 mg·mL-1、4 mg·mL-1、0.0625mg·mL-1、0.0625mg·mL-1,并且B10对于痤疮丙酸杆菌和白色假丝酵母的抑制活性高于盐酸多西环素和氟康唑。
【Abstract】 Using matrine as raw material, 12 hydrazone derivatives of matrine have been designed and synthesized in the presence of phosphorus oxychloride, with yields ranging from 60 to 90%.All compounds were characterized by 1 H-NMR,13 C-NMR and mass spectrometry.The synthesis method is simple in operation, wide in application and easy in post-processing.The antibacterial activity of 15 matrine hydrazone derivatives against two gram-positive bacteria, gram-negative bacteria and one fungus was detected by double dilution method in the hope of finding drug candidates with better antibacterial activity.The results showed that most of the target compounds had different degrees of antibacterial activity, among which B4 and B10 had better antibacterial effect, and their MIC values against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, propionibacter acnes and Candida albicans were B4:1 mg·mL-1,1 mg·mL-1,0.5 mg·mL-1,0.25 mg·mL-1,0.25 mg·mL-1;B10:2 mg/mL,4 mg·mL-1,4 mg·mL-1,0.0625 mg·mL-1,0.0625 mg·mL-1,B10 had higher inhibitory activity against propionibacterium acnei and Candida albicans than doxycycline hydrochloride and fluconazole.
