不同品种藜麦的RAPD分析

【摘要】 藜麦蛋白含量高,营养价值丰富,受到国内外学者的广泛关注。为进一步研究藜麦种质资源的遗传特性,本实验以9个藜麦品种为材料,采用46个随机引物对不同藜麦品种的基因组DNA进行RAPD分析。实验结果表明:通过PCR扩增,一共获得了372条扩增条带,其中多态性条带134条,多态性比例为36%。由聚类分析发现9个藜麦品种之间的遗传相似系数在0.530~0.804之间。同时,9个藜麦品种可以划分为4类。第一类为QA13-8和SCL;第二类为QA13-13-1-23,它与其他品种的遗传距离较远;第三类为黄藜1号和土黄藜;第四类为红藜、黄藜、YY-6-6-Z、QA13-8-1-15。

【Abstract】 Quinoa has high protein content and rich nutritional value, which has been widely studied by scholars. In order to understand the genetic characteristics of quinoa germplasm resources, nine different varieties of quinoa were taken as experimental materials. We used 46 random primers to perform RAPD analysis on the genomic DNA of different quinoa varieties. Through PCR amplification, we obtained 372 bands, among which 134 were polymorphic, with a polymorphism ratio of 36%. According to the clustering analysis, the genetic similarity coffficient of these varieties was between 0.530 and 0.804. These nine quinoa varieties can be grouped into 4 categories. The first category includes QA13-8 and SCL.The second group is QA13-13-1-23, which is genetically distant from other breeds. Yellow quinoa1 and yellowish-brown quinoa are the third category. The last category includes red quinoa, yellow quinoa, YY-6-6-Z and QA13-8-1-15.
