重组弓形虫GRA7的纳米金免疫传感器检测弓形虫GRA7抗体
Detection of antibodies to GRA7 of Toxoplasma gondii by gold nanorod immunosensor based on recombinant Toxoplasma gondii GRA7
【摘要】 目的:建立基于重组弓形虫致密颗粒蛋白7(GRA7)的纳米金免疫传感器检测血清GRA7抗体,评价其检测性能。方法:体外合成gra7基因,构建重组表达质粒pQE-60-gra7,表达并纯化GRA7,Western blot鉴定其免疫活性;种子生长法制备金纳米棒,构建GRA7的纳米金免疫传感器检测人血清GRA7抗体,评价其灵敏度和特异度,并计算阳性、阴性预测值以及诊断效率。结果:成功构建重组表达质粒pQE-60-gra7,纯化GRA7显示良好的反应原性;GRA7的纳米金免疫传感器检测人血清弓形虫GRA7抗体的灵敏度、特异度、阳性预测值、阴性预测值和诊断效率分别为73.21%、100%、100%、77.94%、86.24%。结论:重组弓形虫GRA7的纳米金免疫传感器可用于弓形虫GRA7抗体的检测。
【Abstract】 Objective To establish a gold nanorod immunosensor using recombinant dense granule protein 7(GRA7) of Toxoplasma gondii for detection of antibodies to GRA7 of Toxoplasma gondii,and evaluate its detection performance.Methods Gene of gra7 was synthesized in vitro to construct the recombinant expression plasmid pQE-60-gra7.GRA7 expressed in E.coli M15 containing pQE-60-gra7 was purified by Ni2+-chelating chromatography;Gold nanorods were prepared by seed growth method,and the gold nanorod immunosensor of recombinant GRA7 was constructed to detect antibodies to GRA7 of Toxoplasma gondii in human serum.The sensitivity,specificity,the positive and negative predictive values and diagnostic efficiency of this method were calculated.Results The recombinant expression plasmid pQE-60-gra7 was successfully constructed,and the purified GRA7 was shown with good immunoreactivity;The sensitivity,specificity,positive predictive value,negative predictive value and diagnostic efficiency were 73.21%,100%,100%,77.94% and 86.24%,respectively.Conclusion The gold nanorod immunosensor based on recombinant GRA7 of Toxoplasma gondii can be used for the detection of antibodies of Toxoplasma gondii.
