子痫前期不同病情患者USP22与Th1/Th22/Th17水平变化及相关性研究
Changes and Correlation of USP22 and Th1/Th22/Th17 Levels in Patients with Different Conditions of Preeclampsia
【摘要】 目的探讨子痫前期(preeclampsia, PE)不同病情患者泛素特异性水解酶22(ubiquitin-specific protease 22,USP22)与辅助性T细胞1(helper T cell 1,Th1)/Th22/Th17水平变化及关联性。方法选取2020-01/2021-03月,作者医院收治的77例PE患者,其中32例重度PE患者为重度PE组,其余45例非重度PE患者为非重度PE组,并选取同期77例健康孕妇作为对照组。比较各组临床资料、USP22蛋白、Th1、Th22、Th17水平。采用多因素Logistic回归方程分析PE的相关影响因素。采用Pearson分析USP22蛋白与Th1、Th22、Th17关系。结果重度PE组、非重度PE组、对照组的收缩压(systolic blood pressure, SBP)、舒张压(diastolic blood pressure, DBP)、血尿素氮(blood urea nitrogen, BUN)、血肌酐(serum creatinine, Scr)、血小板(platelet, PLT)、天门冬氨酸氨基转移酶(aspartate aminotransferase, AST)、丙氨酸氨基转移酶(alanine aminotransferase, ALT)、肺水肿、心功能衰竭情况比较,差异有统计学意义(P均<0.01)。重度PE组、非重度PE组USP22蛋白均低于对照组,且重度PE组USP22蛋白低于非重度PE组,差异均有统计学意义(P均<0.05)。对照组、非重度PE组、重度PE组Th1、Th22、Th17依次升高,两两比较差异均有统计学意义(P均<0.05)。调整了SBP、DBP、BUN、Scr、PLT、AST、ALT、肺水肿、心功能衰竭后,USP22蛋白、Th1、Th22、Th17仍与PE发生相关(P<0.05);USP22蛋白与Th1、Th22、Th17呈负相关(r值依次为-0.434、-0.548、-0.607,P均<0.001)。结论 Th1/Th22/Th17细胞免疫增强造成及加重PE,USP22蛋白与Th1/Th22/Th17有负向调控关系,有望成为防治PE的一个新靶点。
【Abstract】 Objective To investigate the changes and relevance in the levels of ubiquitin-specific protease 22(USP22) and helper T cell 1(Th1)/Th22/Th17 in patients with different conditions of preeclampsia(PE).Methods A total of 77 PE patients admitted to the authors′ hospital from January 2020 to March 2021 were selected, 32 severe PE patients were in the severe PE group, the remaining 45 non-severe PE patients were in the non-severe PE group, and 77 healthy pregnat women during the same period were selected as a control group. The clinical data, USP22 protein, and Th1, Th22, Th17 levels of each group were compared. The related influencing factors of PE were analyzed by multivariate Logistic regression equation.The relationship between USP22 protein and Th1, Th22, Th17 was analyzed by Pearson analysis.Results The systolic blood pressure(SBP), diastolic blood pressure(DBP), blood urea nitrogen(BUN), serum creatinine(Scr), platelet(PLT), aspartate aminotransferase(AST), alanine aminotransferase(ALT), pulmonary edema and heart failure of the severe PE group, non-severe PE group and control group were compared, and the difference was statistically significant(all P<0.01). The USP22 protein levels of the severe PE group and the non-severe PE group were lower than those of the control group, and the USP22 protein level of the severe PE group was lower than the non-severe PE group, the difference was statistically significant(all P<0.05). The levels of Th1, Th22 and Th17 of the control group, non-severe PE group and severe PE group increased sequentially, and the difference between each two was statistically significant(all P<0.05). After adjusting SBP, DBP, BUN, Scr, PLT, AST, ALT, pulmonary edema and heart failure, USP22 protein, Th1, Th22 and Th17 were still correlated with PE(P<0.05). USP22 protein was negatively correlated with Th1, Th22, Th17(r=-0.434,-0.548,-0.607, all P<0.001).Conclusion Enhancement of Th1/Th22/Th17 cellular immune causes and aggravates PE, USP22 protein has a negative regulation relationship with Th1/Th22/Th17, which is expected to be a new target for the prevention and treatment of PE.
