黄连药对对DM小鼠胃肠及认知能力的影响

【摘要】 探究黄连药对即黄连-半夏对STZ诱导的糖尿病小鼠胃肠功能以及学习记忆能力的影响.将55只SPF级雄性ICR小鼠按照体重随机分为空白组(n=5),胃肠组(0.65 g·kg-1·d-1)(n=10),模型组(n=10),黄连-半夏组(0.65 g·kg-1·d-1)(n=12),二甲双胍组(0.13 g·kg-1·d-1)(n=7).通过腹腔注射150 mg/kg 1%链脲佐菌素柠檬酸钠缓冲液复制糖尿病小鼠模型,通过一般形态学观察评价黄连-半夏对糖尿病小鼠的治疗作用.采用血糖仪监测造模后第3、10、17 d动物血糖值.并且于造模后第3、10、17 d测定小鼠体重.于取材时测定小鼠的胃残留率和肠推进率探究药物对糖尿病小鼠胃肠功能的影响.采用新物体识别实验评价小鼠的学习记忆能力.实验结果为二甲双胍可降低糖尿病小鼠的体重(P<0.05)以及血糖(P<0.01),黄连-半夏配伍可改善糖尿病小鼠毛色变黄以及精神萎靡等一般形态学症状,另外可降低血糖(P>0.05)和降低糖尿病小鼠体重(P>0.05),另外黄连-半夏能改善糖尿病小鼠的胃肠功能但作用不明显(P>0.05).且黄连-半夏不能改善糖尿病小鼠的学习记忆能力(P>0.05).

【Abstract】 This experiment observed the effect of coptis chinensis-pinellia on the gastrointestinal function and learning and memory ability of STZ-induced diabetic mice. 34 mice were randomly divided into diabetes model group, coptis chinensis-pinellia group(0.65 g·kg-1·d-1), metformin group(0.13 g·kg-1·d-1), and blank group. The diabetic mouse model was replicated by intraperitoneal injection of 150 mg/kg 1% streptozotocin sodium citrate buffer, and the therapeutic effect of coptis chinensis on diabetic mice was evaluated by general morphological observation. A blood glucose meter was used to monitor the blood glucose levels of animals on the 3 rd, 10 th, and 17 th days after modeling. The body weight of mice was measured on the 3 rd, 10 th, and 17 th days after modeling. The gastrointestinal function of diabetic mice was evaluated by measuring the gastric residual rate and intestinal propulsion rate of the mice at the time of sampling. A new object recognition experiment was used to evaluate the learning and memory ability of mice. The result of the experiment was that metformin could reduce the body weight(P<0.05) and blood sugar(P<0.01) of diabetic mice. The combination of coptis chinensis and pinellia could improve the general morphological symptoms of diabetic mice such as yellowing and mental depression, and could also lower blood glucose.(P>0.05) and increase the body weight of diabetic mice(P>0.05), but coptis chinensis-pinellia could not improve the gastrointestinal function and learning and memory ability of diabetic mice(P>0.05). Therefore, the compatibility of coptis chinensis could improve the general morphological symptoms such as mental depression and slow movement of diabetic mice, and could also lower blood sugar and reduce the weight of diabetic mice. In addition, Huanglian-Banxia could improve the gastrointestinal function of diabetic mice but the effect was not obvious(P>0.05). And Huanglian-Banxia could not improve the learning and memory ability of diabetic mice.
