不同抗冻保护剂对绵羊精液冷冻保存效果的影响

【摘要】 为了提高优良种公羊利用效率、促进养羊产业良种化进程,筛选符合当前技术条件的绵羊精液冷冻保护稀释液,试验选择中国美利奴羊(新疆军垦型)公羊12只,采用假阴道法采集精液,测定并分析将不同含量甘油、二甲基乙酰胺(DMA)及甘油+DMA添加于绵羊精液冷冻解冻液后精子的活力、活率、直线速率、曲线速率、顶体完整率及DNA完整率等技术指标,探究不同冷冻保护剂及其不同含量对精液冷冻保存效果的影响。结果表明:在所有甘油组、DMA组和甘油+DMA组中,3.0%甘油+2.0%DMA组冷冻解冻后精子质量最高,活力、活率、直线速率、曲线速率、顶体完整率及DNA完整率分别为(52.93±3.86)%、(65.01±2.04)%、(22.22±0.78)%、(58.71±0.46)%、(50.43±3.78)%、(49.77±1.38)%,2.5%甘油+2.5%DMA组次之。说明在绵羊精液冷冻保护剂中添加3.0%甘油+2.0%DMA的效果优于单独添加甘油和DMA。

【Abstract】 In order to improve the utilization efficiency of excellent rams, promote the process of improved breeding in sheep breeding industry, and screen sheep semen cryopreservation dilutions that meet the current technical conditions, this experiment uses 12 Chinese Merino sheep(Xinjiang Junken type) rams as the test subjects. Semen of ram was collected by pseudovagina method. Then the motility, viability, linear velocity, curve velocity, acrosome integrity and DNA integrity of sheep semen were measured after freezing and thawing using glycerol, dimethylacetamide(DMA) and glycerol plus DMA. The effect of different concentrations of cryoprotectant on the cryopreservation of semen was investigated. The results showed that: among all glycerol groups, DMA groups and glycerol+DMA groups, 3.0% glycerol+2.0% DMA group had the highest sperm quality after freezing and thawing. The motility the viability, linear velocity, curve velocity, acrosome integrity and DNA integrity were(52.93±3.86)%,(65.01±2.04)%,(22.22±0.78)%,(58.71±0.46)%,(50.43±3.78)%,(49.77±1.38)%, followed by the 2.5% glycerol+2.5%DMA group. It shows that the effect of adding 3% glycerol + 2% DMA to sheep semen cryoprotectant is better than adding glycerol and DMA alone.
