β-细辛醚对AD小鼠脑中Aβ沉积的影响

【摘要】 石菖蒲为天南星科植物石菖蒲(Acorus tatarinowii Schott)的干燥根茎,为常用中药,有开窍豁痰、醒神益智等功效。药理学研究表明β-细辛醚是石菖蒲中的主要活性成分。文章研究β-细辛醚对阿尔兹海默症(Alzheimer’s disease, AD)小鼠脑中Aβ沉积的影响。研究采用5×FAD转基因小鼠,由4月龄开始给予β-细辛醚灌胃,每天剂量为10 mg/kg,持续90 d。灌胃结束后,对小鼠脑组织进行冰冻切片,采用刚果红和组织免疫染色鉴定AD小鼠脑中老年斑的沉积情况。结果表明,β-细辛醚组AD小鼠脑中Aβ沉积斑块的数量显著减少,表明β-细辛醚在可减少5×FAD转基因小鼠老年斑的生成,改善AD的特征性病理表现。

【Abstract】 β-asarone is the main active ingredient of Acorus tatarinowii Schott, one of the commonly used Chinese herbs for dementia. This study aims to investigate whether β-asarone could improve the β-amyloid(Aβ) deposition in the brain of mice with Alzheimer’s disease(AD). The 5×FAD mice were used in this study. From the age of four months, the mice in the experimental group were given β-asarone, and the dose of each mice was 10 mg/kg per day for 90 days. After intragastric administration, the mice brain was collected for frozen section, and Congo red staining and tissue immune staining were used to identify the deposition of senile plaques(SPs) in the brain of AD mice. The results showed that the number of Aβ deposited plaques in the brain of AD mice was significantly reduced, indicating that β-asarone could inhibit the formation of SPs of 5×FAD mice and improve the characteristic pathological changes of AD.
