一株磺胺二甲嘧啶降解菌的筛选、鉴定及生长特性研究
Screening,Identification and Growth Characteristics of a Sulfadimidine Degrading Strain
【摘要】 磺胺二甲嘧啶(SMZ)是一种结构复杂、化学性质稳定且应用广泛的抗生素,因使用过量在各种环境尤其是地下水中被频繁检出.采用微生物治理手段,培养SMZ高效降解菌株是解决地下水中SMZ污染问题的有效方法.采用稀释涂布平板法从北京市某区域地下水中筛选出一株SMZ降解菌株,经形态学观察、生理生化实验及16S rRNA分子鉴定,鉴定其为斯氏假单胞菌,命名为D3菌株,并探究其生长特性.实验结果表明温度、pH、接种量、SMZ浓度均会影响D3菌株的生长,且温度为25℃,pH为6、SMZ浓度为5 mg/L,接种量为1%的条件是其最适生长条件.该研究成果为进一步解析其降解机制提供研究基础和数据支持,为SMZ高效降解菌优质菌种的开发提供资源.
【Abstract】 Sulfadimethamine(SMZ) is a kind of antibiotic with complex structure, stable chemical properties and wide application. Because of excessive use, SMZ is frequently detected in various environments, especially groundwater. It is an effective method to solve the problem of SMZ pollution in groundwater by microorganism treatment and cultivation of SMZ efficient degradation strains. In this study, a SMZ-degrading strain was screened from groundwater in a certain area of Beijing by dilution coating plate method. After morphological observation, physiological and biochemical experiments and 16 S rRNA molecular identification, it was identified as Pseudomonas Skandii, named strain D3, and its growth characteristics were investigated. The results showed that temperature, pH, inoculum size and SMZ concentration all affected the growth of D3 strain, and the optimal growth conditions were 25℃, pH 6, SMZ concentration 5 mg/L and inoculum size 1%. The research basis and data support for further analysis of the degradation mechanism of SMZ, and the resources for the development of high quality strains of SMZ efficient degradation strain are provided.
