人参四逆颗粒对慢性心力衰竭大鼠心功能和神经内分泌因子水平的影响

【摘要】 目的 探讨人参四逆颗粒对慢性心力衰竭大鼠心功能和神经内分泌因子水平的影响。方法 在90只SD大鼠中随机选取30只设为空白对照组,剩余60只大鼠采用腹腔注射盐酸阿霉素的方法建立慢性心力衰竭模型。建模成功后将60只大鼠随机分为模型组和给药组各30只。空白对照组与模型组大鼠均给予常规饲养及生理盐水灌胃,给药组大鼠给予中等剂量(0.5 g/kg)人参四逆颗粒灌胃。干预21 d后,采用心脏超声诊断仪测定3组大鼠的心功能指标,并采用ELISA法检测血浆B型钠尿肽(BNP)、内皮素、血管紧张素Ⅰ(AngⅠ)、血管紧张素Ⅱ(AngⅡ)水平。结果 与空白对照组相比,模型组左室舒张末期室间隔厚度、左室后壁厚度、左室舒张末期内径、左室收缩末期内径、左心室重量、左室舒张末期容积均增大,而左室射血分数降低;而给药组以上心功能指标均优于模型组(均P<0.05)。模型组血浆BNP、内皮素、AngⅠ、AngⅡ水平均高于空白对照组,而给药组以上指标水平均低于模型组(均P<0.05)。结论 人参四逆颗粒能改善慢性心力衰竭大鼠的心脏功能,其或通过降低神经内分泌因子水平而发挥治疗慢性心力衰竭的作用。

【Abstract】 Objective To investigate the effect of Renshen Sini Granules on cardiac function and neuroendocrine factors levels in rats with chronic heart failure. Methods Thirty out of 90 SD rats were randomly selected as blank control group, and the remaining 60 rats were intraperitoneally injected with doxorubicin hydrochloride to establish the models of chronic heart failure. After successful modeling, the 60 rats were randomly divided into model group(n=30) and medication group(n=30). Rats in the blank control and model groups were conventionally reared and intragastrically given normal saline, while rats in the medication group were intragastrically given Renshen Sini Granules at a moderate dose(0.5 g/kg). Twenty-one days after the intervention, echocardiography diagnosis apparatus was utilized to determine the cardiac function indicators of rats in the three groups, and ELISA assay to detect the plasma levels of B-type natriuretic peptide(BNP), endothelin, angiotensin Ⅰ(AngⅠ), and angiotensin Ⅱ(AngⅡ). Results Compared with the blank control group, the model group yielded increases in left ventricular end-diastolic interventricular septal thickness, left ventricular posterior wall thickness, left ventricular end-diastolic diameter, left ventricular end-systolic diameter, left ventricular mass, and left ventricular end-diastolic volume, and a decrease in left ventricular ejection fraction; furthermore, the medication group was superior to the model group in the above cardiac function indicators(all P<0.05). The model group reported higher plasma BNP, endothelin, AngⅠ, and AngⅡ levels than the blank control group, whereas the levels of the above indicators were lower in the medication group than in the model group(all P<0.05).Conclusion Renshen Sini Granules can improve the cardiac function of rats with chronic heart failure, which may play a role in the treatment of chronic heart failure by reducing the levels of neuroendocrine factors.
