培养组学联合MALDI-TOF MS技术对医院环境中昆虫标本的菌群分析

【摘要】 目的 通过培养法和基质辅助激光解吸电离飞行时间质谱仪(MALDI-TOF MS)对昆虫标本菌群进行分析,探讨两者联合使用的价值,以及昆虫标本菌群特征和对抗菌药物的敏感性。方法 采集医院环境中的昆虫标本,对标本进行碾磨和进行梯度稀释,然后接种于培养平板,培养、分离标本中的单个菌落,利用MALDI-TOF MS进行菌种鉴定。对分离出来的肠杆菌目细菌采用微量肉汤稀释法进行药敏试验。结果 该研究从16只昆虫标本中共分离出104株菌,6株菌无法通过MALDI-TOF MS进行鉴定,MALDI-TOF MS的鉴定成功率为94.23%,平均鉴定分值为2.075。MALDI-TOF MS成功鉴定的98株菌中,革兰阳性菌56株(57.14%),革兰阴性菌41株(41.84%)。革兰阳性菌中,芽孢杆菌属分离率最高,共30株,占53.57%。革兰阴性菌中以肠杆菌目细菌为主,共分离29株(70.73%),分离株对抗菌药物的敏感性较高。结论 培养组学联合MALDI-TOF MS可以用于昆虫标本中菌株鉴定。由于昆虫的流动性,其携带的菌株可能成为临床潜在的感染病原体。

【Abstract】 Objective To analyze the microbiota of insect specimens by culture method and matrix-assisted laser desorption/ionization-time of flight mass spectrometry(MALDI-TOF MS),and the value of the combined use of the two, as well as the characteristics of the microbiota of insect specimens and their susceptibility to antibiotics.Methods Collect insect specimens in the hospital environment.The samples were milled and serially diluted, then plated on culture plates.Single colonies were cultured and isolated, and MALDI-TOF MS was used for bacterial species identification.Antimicrobial susceptibility of isolated Enterobacteriaceae was tested by micro-dilution method.Results In this study, a total of 104 strains of microbial colonies were isolated from 16 insect specimens, and 6 strains could not be identified by MALDI-TOF MS.The success rate of identification by using MALDI-TOF MS was 94.23%,and the average identification score was 2.075.Among the 98 strains which were successfully identified by MALDI-TOF MS,56 strains(57.14%) of Gram-positive strains and 41 strains(41.84%) of Gram-negative strains were identified.Among the Gram-positive bacteria, Bacillus genus had the highest isolation rate, with a total of 30 strains, accounting for 53.57%.The Gram-negative bacteria were dominated by Enterobacteriaceae, and 29 strains(70.73%) were isolated, and the isolates were highly sensitive to antibiotics.Conclusion Culturomics combined with MALDI-TOF MS can be used for strain identification in insect specimens.Due to the mobility of insects, the strains carried by them may become clinical potential infectious pathogens.
