MFAP5在非小细胞肺癌组织中的表达情况及其对癌细胞增殖和转移的影响

Expression and significance of MFAP5 in non-small cell lung cancer and its effect on cancer cell proliferation and metastasis

【摘要】 目的 探讨MFAP5在非小细胞肺癌(NSCLC)疾病进展中的作用。方法 收集NSCLC患者的癌组织及其对应的癌旁组织各87例,免疫组化检测组织中MFAP5表达。体外培养人NSCLC细胞A549和人正常肺上皮细胞BEAS-2B,检测两种细胞中MFAP5的表达水平。进一步用慢病毒液感染部分A549细胞,分为sh-NC组和sh-MFAP5组,不进行病毒感染操作的A549细胞作为空白对照组,检测敲低MFAP5后对NSCLC细胞的影响。实时荧光定量反转录PCR(qRT-PCR)检测细胞MFAP5 mRNA表达;Western blot检测细胞MFAP5、神经型钙黏蛋白(N-cadherin)、上皮型钙黏蛋白(E-cadherin)和波形蛋白(Vimentin)蛋白表达;CCK-8检测细胞活力;克隆形成实验检测细胞增殖;细胞划痕实验检测细胞迁移;Transwell实验检测细胞侵袭。结果 与癌旁组织相比,NSCLC组织中MFAP5 IHC评分和MFAP5高表达率明显升高(P<0.05)。NSCLC组织中MFAP5表达水平与患者淋巴结转移、TNM分期有关(P<0.05),而与患者年龄、性别、吸烟史和组织学类型无关(P>0.05)。与BEAS-2B细胞相比,A549细胞中MFAP5 mRNA和蛋白表达明显升高(P<0.05)。与空白对照组相比,sh-NC组细胞中各指标差异无统计学意义(P>0.05);与sh-NC组相比,sh-MFAP5组细胞中MFAP5 mRNA和蛋白表达以及细胞活力、增殖能力、划痕愈合率和侵袭细胞数明显降低(P<0.05),N-cadherin和Vimentin表达明显降低(P<0.05),E-cadherin表达明显升高(P<0.05)。结论 MFAP5在NSCLC组织中高表达,与NSCLC淋巴结转移、TNM分期有关,参与调控NSCLC细胞增殖和转移。

【Abstract】 Objective To explore the role of microfibril-associated protein 5(MFAP5) in the progression of non-small cell lung cancer(NSCLC).Methods A total of 87 cases of cancerous tissues and corresponding adjacent tissues of NSCLC patients were collected.Immunohistochemistry was used to detect the expression of MFAP5,N-cadherin, E-cadherin, Vimentin in the tissues.The human non-small cell lung cancer cells A549 and human normal lung epithelial cells BEAS-2 B were cultured in vitro and the expression level of MFAP5 in the two types of cells were detected.Further, some A549 cells were infected with lentivirus solution and divided into sh-NC group and sh-MFAP5 group to detect the effect of knockdown of MFAP5 on NSCLC cells.qRT-PCR was used to detect the expression of MFAP5 mRNA in cells.Western blot was used to detect the expression of MFAP5 N-cadherin, E-cadherin, Vimentin protein in cells.CCK-8 was used to detect cell viability.The formation test was used to detect cell proliferation, the cell scratch test was used to detect the migration of cells, the Transwell test was used to detect the invasion of cells.Results Compared with tissues adjacent to cancer, the MFAP5 IHC score and the high expression rate of MFAP5 in NSCLC tissues were significantly higher(P<0.05).The expression level of MFAP5 in NSCLC tissue was related to lymph node metastasis and TNM staging(P<0.05),but not related to the patients′ age, gender, smoking history and histological type(P>0.05).Compared with the BEAS-2 B cells, the expression of MFAP5 mRNA and protein in the A549 cells was significantly increased(P<0.05).Compared with the blank control group, there was no statistically significant difference in the indicators in the sh-NC group(P>0.05).Compared with the sh-NC group, the expression of MFAP5 mRNA and protein, cell viability, proliferation capacity, scratch healing rate, and number of invasive cells in the sh-MFAP5 group were significantly reduced(P<0.05),and the expression of N-cadherin and Vimentin proteins significant decreased(P<0.05),the expression of E-cadherin protein was significantly increased(P<0.05).Conclusion MFAP5 is highly expressed in NSCLC tissues, which is related to NSCLC lymph node metastasis and TNM staging, and participates in regulating the proliferation and metastasis of NSCLC cells.
