成纤维细胞生长因子受体3对大鼠失神经骨折愈合的影响
The mechanism of FGFR3 on the healing of denervation fracture in rats
【摘要】 目的:探索成纤维细胞生长因子受体3(FGFR3)在失神经骨折愈合中的调节作用和具体机制。方法:将36只大鼠随机分为实验组和对照组,每组18只。所有大鼠建立T10脊髓横断合并胫骨骨折动物模型,术前实验组大鼠给予腹腔注射FGFR3阻滞剂PD173074,对照组注射等量生理盐水。术后采用TARLOR评分,评价实验大鼠神经功能;术后7、21天拍摄X线片,观察骨折愈合情况,同时行Masson三色法染色,定量分析骨痂纤维化程度。结果:TARLOR评分显示上述两组大鼠均达到实验要求。X线片提示实验组大鼠骨折术后较对照组愈合速度快,骨痂成熟程度高,同时Masson三色法染色提示实验组骨折修复结果较对照组好,两组大鼠胫骨组织纤维率亦有明显差别(P<0.05)。结论:FGFR3低表达促进失神经骨折愈合的进程,加速了骨痂的塑形,在机体骨组织生长发育与骨折愈合重建中发挥着重要的作用。
【Abstract】 Objective:We explore the role and specific mechanism of FGFR3 in the denervation fracture. Methods:In this study,36 rats were randomized into experimental group and control group,18 each.All of these rats were established animal model of T10 spinal cord injury(SCI)with tibial fracture state,then the experimental group was given FGFR3 blocker PD173074 by intraperitoneal injection,while equivalent saline was given in the control group before surgery.Tarlor score was used to evaluate nerve function of all the rats after surgery;X-ray was taken at 7 and 21 days to observe fracture healing after surgery;Masson trichrome staining measured the fiber rate of the tibia at the same time. Results:The Tarlor behavioral scores showed both groups of rats met the experimental requirements. X-ray indicated that fracture in the experimental group healed faster,there were also significant differences in tibial fiber rate determination in the two groups(P<0.05). Conclusions:FGFR3 low expression promotes the healing process of denervation fracture and plays an important role in the growth of bone tissue and the reconstruction of fracture healing.
