木榄耐盐内生细菌分离及其16S rDNA鉴定
Isolation and 16S rDNA Identification of Salt-tolerant Bacteria of Bruguiera gymnorhiza
【摘要】 [目的]从木榄根茎叶中分离筛选鉴定耐盐碱菌株。[方法]以不同盐浓度的LB培养基分离筛选木榄根茎叶中耐盐内生细菌,对其进行生理生化分析和分子鉴定。[结果]从根茎叶中初步分离出耐盐的内生细菌24株,经革兰氏染色、过氧化氢试验、吲哚试验与甲基红试验均为阳性,淀粉水解酶有1株细菌为阴性。提取各菌株的DNA并扩增其16S rD NA序列,以Hinf Ⅰ等4种酶进行酶切,选择不同类型酶切结果的代表菌株测序,BLAST比对分析并利用MEGA软件建立发育树,初步推断出,所分离的内生细菌分别为越南芽孢杆菌、蜡样芽孢杆菌、白翎芽孢杆菌和巴氏葡萄球菌。[结论]该研究为培育耐盐农作物提供了行之有效的技术理论基础。
【Abstract】 [Objective] The isolation,screening and identification of saline-alkali resistant strains from the roots and leaves of Olive were studied. [Method] Salt-tolerant endophytic bacteria from roots and leaves of Bruguiera gymnorrhiza were isolated and screened by LB medium with different salt concentrations,and their physiological and biochemical analysis and molecular identification were carried out. [Result] The results showed that 24 strains of salt-tolerant endophytic bacteria were isolated from the roots,stems and leaves. Gram staining,hydrogen peroxide test,indole test and methyl red test were all positive,starch hydrolase was negative in one bacterium. The DNA of each strain was extracted and its 16S rDNA sequence was amplified,and four kinds of enzymes,such as Hinf Ⅰ,were used for digestion. The representative strains with different types of digestion results were selected for sequencing,and BLAST comparative analysis was carried out,and the phylogenetic tree was established by using MEGA software to preliminarily deduce,the isolated endophytic bacteria are respectively Bacillus vietnamensis,Bacillus cereus,Bacillus baekryungensis,and Staphylococcus pasteuri. [Conclusion] This study provided an effective technical and theoretical basis for cultivating salttolerant crops.
