花生红衣水提物对对氯苯丙氨酸失眠模型大鼠神经递质的影响

【摘要】 目的 观察花生红衣水提物对对氯苯丙氨酸(PCPA)所致失眠模型大鼠的治疗作用并探讨其可能的作用机制。方法 60只SD雄性大鼠,随机分为正常对照组、模型对照组、地西泮阳性对照组、花生红衣水提取物高、中、低剂量(650、325、162.5 mg/kg)组,每组10只。除正常对照组外,其余各组均采用腹腔注射400 mg/kg对氯苯丙氨酸(PCPA)混悬液,制备失眠大鼠模型,连续注射3 d,大鼠昼夜节律消失,模型制备成功。造模成功后花生红衣水提物高、中、低剂量组,阳性药组,按照1 mL/100 g体重灌胃给药,正常对照组与模型对照组给予等体积生理盐水灌胃,连续7 d,观察记录各组大鼠实验期间睡眠潜伏期、睡眠时间和体质量的变化。ELISA法测定各组大鼠下丘脑中神经递质5-羟色胺(5-HT)、褪黑素(MT)的含量。结果 与正常对照组相比,模型组大鼠睡眠潜伏期明显延长(P<0.05),睡眠时间显著缩短(P<0.05),脑组织中5-HT和MT含量均明显减少(P<0.05)。与模型组相比,花生红衣水提取物(高、中、低)剂量组和阳性对照组大鼠,睡眠潜伏期明显缩短(P<0.05),睡眠时间显著延长(P<0.05);可显著升高脑组织中5-HT和MT的含量(P<0.05)。结论 花生红衣水提取物可改善由PCPA引起的失眠,其作用机制可能与提高大鼠下丘脑组织中5-HT和MT的含量有关,为花生红衣水提物治疗失眠提供了实验依据。

【Abstract】 Objective To evaluate the therapeutic effect and mechanism of peanut red skin water extract on P-chlorophenylalanine(PCPA)induced insomnia model rats. Methods All 60 male SD rats were randomly divided into the normal control group,the model group,the Diazepam positive control group,and high,middle and low dose of peanut red skin aqueous extract groups(650 mg/kg,325 mg/kg,162.5 mg/kg). 10 rats in each group. In addition to the normal control group,each group was intraperitoneally injected with 400 mg/kg PCPA suspension with 1 mL/100 g to replicate the model of insomniac rats,and the normal group was given the same volume of normal saline,which was continuously injected for 3 d,The disappearance of circadian rhythm in rats indicated that the model was successfully prepared. After modeling,the high,middle,low-dose of peanut red skin aqueous extract groups and the Diazepam positive group were administered orally at1 mL/100 g,while the normal control group and the model group were given normal saline of the same volume for 7 d continuously. The sleep latency,sleep time and the changes of body weight of rats in each group were observed and recorded. Content changes of 5-HT and MT of rats in each group were measured by ELISA. Results Compared with the normal control group,the sleep latency of model group was significantly prolonged(P<0.05),and the sleep time was significantly shortened(P<0.05). the contents of 5-HT,MT in brain tissue were significantly decreased(P<0.05). Compared with the model group,the sleep latency was significantly shortened(P<0.05)and the sleep time was significantly prolonged(P< 0.05)in peanut red skin water extract(high,medium and low)dose groups and positive control group. The contents of 5-HT and MT in brain tissue were also significantly increased(P<0.05). Conclusion The peanut red skin water extract can reverse insomnia caused by PCPA,Its mechanism may be related to the increase of 5-HT and MT content in the hypothalamus of rats.The result provide experimental basis for the treatment of insomnia by the peanut red skin water extract.
