基于网络药理学探讨双黄连颗粒治疗新型冠状病毒肺炎的可行性

【摘要】 目的:通过网络药理学方法探析双黄连颗粒治疗新型冠状病毒肺炎(COVID-19)的活性成分、靶点和信号通路,阐明其潜在分子机制。方法:借助中药系统药理学数据库与分析平台检索双黄连颗粒组成药材的活性成分和潜在靶点,GeneCards数据库中获取疾病靶点。使用Cytoscape构建中药-化合物-靶点信息网络。利用String构建蛋白质相互作用(PPI)网络,Metascape数据库进行富集分析。结果:筛选后得到双黄连颗粒活性成分68个,靶点253个。信息网络显示槲皮素、山柰酚、木犀草素等发挥关键作用。PPI分析可知双黄连颗粒主要通过AKT1、ALB、TP53等靶点发挥作用。KEGG通路富集到病毒相关通路、PI3K-Akt信号通路等。结论:双黄连颗粒作用于不同靶点、多条信号通路发挥抗病毒、抗炎等作用,实现对COVID-19的治疗作用。

【Abstract】 Objective: To explore the active compounds, targets and signal pathways of Shuanghuanglian Granules in the treatment of new coronavirus pneumonia(COVID-19) by network pharmacology, and elaborate its potential molecular mechanism. Methods: The active compounds and potential targets of Shuanghuanglian granules were retrieved from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform. The COVID-19 targets were screened through the GeneCards. Cytoscape software was used to build a Chinese material medica-compound-target network. A protein-protein interaction(PPI)network was built by the String database. Metascape database was used to perform the enrichment analysis. Results: Shuanghuanglian granules contained 68 active compounds and 253 targets. The quercetin, kaempferol, luteolin and so on played a key role in the network. Shuanghuanglian mainly exert their effects through AKT1, ALB, TP53 and so on. KEGG pathway enrichment analysis shows virus-related pathways, PI3 K-Akt signaling pathway and so on. Conclusion: The active compounds of Shuanghuanglian granules work on different targets and participate in multiple signal pathways. They exert anti-viral, anti-inflammatory effects,realizing the therapeutic effect on COVID-19.
