常压微波辅助法制备高荧光量子产率水溶性硅纳米粒子及其在细胞荧光成像中的应用

【摘要】 将高压微波辅助合成方法改进为常压微波辅助法,在常压条件下,以N-(2-氨基乙基)-3-氨基丙基三乙氧基硅烷为新的硅源,柠檬酸钠为还原剂,丙三醇为溶剂,合成了高荧光量子产率的水溶性硅纳米粒子(SiNPs)。其在发射波长500 nm处发射明亮的蓝色荧光,荧光量子产率达45.6%。SiNPs具有优异的pH稳定性和耐盐性。细胞毒性实验结果表明,SiNPs具有较低的生物毒性和良好的生物相容性。将SiNPs应用于HeLa细胞的荧光成像,效果较好。

【Abstract】 The high pressure microwave-assisted synthesis method was improved to atmospheric pressure microwave-assisted synthesis. Using N-(2-aminoethyl)-3-aminopropyl triethoxysilane as a new silicon source,sodium citrate as reducing agent,and glycerol as solvent,water-soluble silicon nanoparticles(SiNPs)with high fluorescence quantum yield were synthesized under atmospheric pressure. The synthesized SiNPs emitted bright blue fluorescence at 500 nm,and the fluorescence quantum yield was as high as 45.6%. Meanwhile,the prepared SiNPs had excellent pH stability and good resistance to salt solution. The results of cytotoxicity experiments showed that the synthesized SiNPs had low toxicity and good biocompatibility. Finally,the obtained SiNPs were successfully applied to the fluorescence imaging of HeLa cells.
