FAdV-4感染对白羽肉鸡外周血细胞、血液生化指标和炎症因子的影响
Effects of FAdV-4 infection on peripheral blood cells, blood biochemistry and inflammatory factors in white feather broilers
【摘要】 采用Ⅰ群禽腺病毒血清4型(FAdV-4)人工感染白羽肉鸡,于感染3、6、12、24 h时检测并分析FAdV-4对肉鸡外周血细胞、血液生化指标和炎症因子的影响.结果表明,3~24 h时,被FAdV-4感染的时间越长,肉鸡外周血细胞病毒载量越高.对于外周血细胞,肉鸡红细胞数与病毒载量呈负相关,感染6 h时较未攻毒组显著减少(P<0.05);白细胞数与病毒载量呈正相关,感染6 h时较未攻毒组显著增加(P<0.05);感染12、24 h时,血小板数较未攻毒组显著增加(P<0.05).对于血液生化指标,感染3~24 h时,肉鸡血液中的血糖浓度较未攻毒组显著升高(P<0.05),甘油三酯浓度较未攻毒组显著下降(P<0.05);感染6、24 h时,总蛋白含量较未攻毒组显著下降(P<0.05).对于炎症因子,感染3~24 h时,肉鸡外周血中组胺和缓激肽的含量较未攻毒组显著增加(P<0.05);感染6 h时,白介素-1β(IL-1β)、白介素-6(IL-6)和P物质的含量与未攻毒组的差异不显著(P>0.05),感染3、12、24 h时较未攻毒组显著增加(P<0.05);人高迁移率族蛋白B1(HMGB1)只在感染24 h时较未攻毒组显著增加(P<0.05).可见,感染FAdV-4后,白羽肉鸡的红细胞数、血脂浓度和总蛋白含量下降,白细胞数和血糖浓度升高,极大地促进外周血炎症因子的释放,产生了严重的炎症反应.
【Abstract】 White feather broilers were infected with Fowl adenovirus serotype 4(FAdV-4), then the peripheral blood cell, biochemical index and inflammatory factor were detected at 3, 6, 12 and 24 h after the infection. The results showed that the viral load of peripheral blood increased with the time after infection. The number of erythrocytes was negatively correlated with the viral load, and was significantly reduced(P<0.05) at 6 h after the infection. On the contrary, the number of white blood cells was positively corelated with the viral load, and was significantly increased(P<0.05) at 6 h. The number of platelets was significantly elevated(P<0.05) at 12 and 24 h post infection. For blood biochemical indexes, the glucose levels were significantly increased(P<0.05), but the triglycerides levels were significantly reduced after the infection(P<0.05), and the total protein levels were decreased significantly at 6 and 24 h(P<0.05). The contents of histamine and bradykinin were significantly increased after the infection(P<0.05). As such, the contents of interleukin-1β(IL-1β), interleukin-6(IL-6) and substance P were significantly increased(P<0.05) after the infection with the exception at 6 h without significant difference. The human high-mobility group box-1(HMGB1) was significantly increased at 24 h(P<0.05). To summarize, FAdV-4 infection led to lower levels of erythrocytes, blood lipids and total protein while higher levels of white blood cell and glucose, which were contributed to the releases of inflammatory factors and subsequent severe inflammatory responses.
