珍珠芦荟组织培养技术
Tissue Culture of Aloe aristate
【摘要】 为了提高珍珠芦荟种苗繁殖效率,于2020年在漳州市农业科学研究所,以健康幼年珍珠芦荟植株作为材料,进行组织培养快速繁殖技术研究,筛选适合的诱导、继代、生根培养基,同时探索移栽条件对组培苗成活率的影响。结果表明:以MS培养基为基础,添加6-BA 3.0 mg·L-1有利于珍珠芦荟诱导,诱导率达54.3%;在2/3 MS培养基中添加6-BA 3.0 mg·L-1+NAA 1.0 mg·L-1+蔗糖40 g·L-1能促进珍珠芦荟增殖,增殖系数为4.2;在1/2 MS培养基中添加IBA 0.5 mg·L-1+NAA 1.0 mg·L-1+蔗糖20 g·L-1能促进生根,且根系比较健康;珍珠芦荟组培苗经过15 d炼苗,在通风、阴凉处晾5 d后移栽到栽培基质(进口草炭∶赤玉土=3∶1)中,移栽成活率最高。
【Abstract】 In order to improve the propagation efficiency of Aloe aristate,healthy young Aloe aristate were used to explore the technology of rapid propagation of tissue culture; meanwhile, induction, subculture and rooting medium of Aloe aristate were selected, the effect of transplanting conditions on the survival rate of tissue culture seedlings was also explored.The results showed that: for MS medium, adding 3.0 mg·L-1 6-BA was beneficial to the induction of Aloe aristate,and the induction rate was 54.3%;adding 3.0 mg·L-1 6-BA,1.0 mg·L-1 NAA and 40 g·L-1 sugar into 2/3 MS medium could promote the proliferation of Aloe aristate,and the proliferation coefficient was 4.2;adding 0.5 mg·L-1 IBA,1.0 mg·L-1 NAA and 20 g·L-1 sugar into 1/2 MS medium could promote rooting, and the root system was healthy; the survival rate of Aloe aristate plantlets was the highest after 15 days of refining, 5 days of airing in a ventilated and cool place, and transplanting into the cultivation medium(imported Peat: Red Jade soil=3∶1).
