基于转录组数据开发福建柏EST-SSR标记

【摘要】 福建柏是一种优良的材用树种,主要分布于中国东南部至西南部以及越南北部。开发表达序列标签-简单序列重复(Expression sequence tags-simple sequence repeat, EST-SSR)分子标记有利于完善福建柏育种策略。采用转录组测序技术(RNA-seq)进行测序及基于转录组组装的SSR的开发,初步获得109个引物和12个EST-SSR多态性标记。从3个种群的57个福建柏个体中鉴定的等位基因数目为2~8个。观测杂合度(Ho)和期望杂合度(He)的范围分别在0~1和0.051~0.775之间,均值分别为0.277和0.420等位基因多态性信息含量的范围在0.139~0.784之间,平均为0.482。实验结果提供了一套可用于监测福建柏种群动态以及发展育种和遗传保护措施的微卫星标记。

【Abstract】 Fokienia hodginsii(Dunn) Henry et Thomas, a timber tree species, is distributed across southeastern to southwestern China, and northern Vietnam.Development of expression sequence tags-simple sequence repeat(EST-SSR) marker is beneficial to provide optimal protection breeding strategy for F.hodginsii.We prepared transcriptome assembly with RNA sequencing.Based on the SSR mining across the transcriptome assembly, 109 primers were selected for initial confirmation and 12 polymorphic EST-SSR loci were obtained.The allele number identified from 57 F.hodginsii individuals which originating from three different populations were ranged from two to eight per locus.The observed and expected heterozygosity levels were ranged from 0 to 1 and 0.051 to 0.775 with an average of 0.277 and 0.420 respectively.Allelic polymorphism information content was ranged from 0.139 to 0.784 with an average of 0.482.This study provides a set of microsatellite markers available for monitoring population dynamics and employing breeding and conservation genetic measures of F.hodginsii.
