伪狂犬病病毒FJMH1907b株的分离和gD基因的演化分析

【摘要】 为探究福建省新流行的猪伪狂犬病病毒(Pseudorabies virus,PRV)g D基因的遗传进化情况,根据GenBank已公布的PRV gD基因序列设计1对特异性扩增引物,对新分离PRV FJMH1907b株的gD基因进行PCR扩增、测序,与国内外已发表的15个毒株进行同源性比对分析,并构建遗传进化树。新分离FJMH1907b毒株与国外Kaplan毒株及疫苗毒Bartha株不属于一个大的分支上,与国内FJ-Y21株、Anhui-ZJ1株、LC株及TJ株同属一个小分支上;新分离FJMH1907b毒株与其它参考毒株gD蛋白的同源性为87.7%~100%,与国外毒株的同源性为87.7%~99.7%。本研究为丰富福建省猪群PRV的分子流行病学和掌握PRV的分子遗传进化规律奠定基础。

【Abstract】 In order to explore the genetic evolution of the g D gene of the newly epidemic porcine pseudorabies virus(PRV) in Fujian province, a pair of specific amplification primers were designed according to the PRV gD gene sequence published by GenBank. The gD gene of the newly isolated PRV FJMH1907 b strain was amplified and sequenced by PCR, compared with 15 published strains at home and abroad, and the genetic evolution tree was constructed. The results showed that the newly isolated FJMH1907 b strain did not belong to a large branch with foreign Kaplan strain and Bartha strain, but belonged to a small branch with domestic FJ-Y21 strain,Anhui-ZJ1 strain, LC strain and TJ strain; The homology of g D protein between the newly isolated FJMH1907 b strain and other reference strains was 87.7%~100%, and 87.7%~99.7% with foreign strains. This study laid a foundation for enriching the molecular epidemiology of PRV in pigs in Fujian province and mastering the molecular genetic evolution law of PRV.
