硫酸化朱砂七多糖对免疫应激鸡下丘脑与肾上腺c-fos表达及血清皮质醇含量的影响

【摘要】 目前中药多糖在动物养殖生产中的应用日益广泛,对于其作用机理的探讨十分必要。以雏鸡为试验用动物,以制备的朱砂七多糖(Polygonum cillinerve polysaccharide, PCP)和硫酸化朱砂七多糖(sulfated Polygonum ciliinerve polysaccharides, sPCP)为材料,将雏鸡随机分为对照组,新城疫(ND)疫苗组,高、低PCP组和高、低sPCP组,在疫苗首免后7 d(21日龄)和二免后7 d(35日龄)分别进行各组鸡的血样采集,制备血清;并取各组鸡的下丘脑和肾上腺制备石蜡切片,进行HE染色和c-fos免疫组化染色;ELISA法测定各组雏鸡血清中皮质醇的含量,观察朱砂七多糖和硫酸化朱砂七多糖对ND疫苗免疫应激鸡的保护作用。结果显示,sPCP组及PCP组雏鸡下丘脑室旁核和肾上腺皮质的c-fos阳性反应均较疫苗组的弱,以低剂量sPCP组的阳性反应最弱;在D21,sPCP低剂量组血清皮质醇含量较疫苗组明显降低,在D35,sPCP低剂量组及PCP组的血清皮质醇含量均低于疫苗组的,但差异不显著(P>0.05)。以上结果说明PCP和sPCP具有一定的抗免疫应激作用,且以低剂量sPCP效果较好。

【Abstract】 Chinese herbs are widely used in the animal husbandry and veterinary medicine.In this study, chickens were used as the experimental animal to detect the protective effect of PCP(Polygonum cillinerve polysaccharide)and sPCP(sulfated Polygonum ciliinerve polysaccharides) on immune stress chickens.Chickens were randomly divided into control group, vaccine groups, PCP high/low-dose group and sPCP high/low-dose groups.Sera were collected at the seventh day after the first immunization(21-day old) and the seventh day after the second immunization(35-day old).Paraffin sections of hypothalamus and adrenals were prepared and stained with hematoxylin-eosin and c-fos immunohistochemistry.Enzyme-linked immunosorbent assay(ELISA) were used to study the levels of serum cortisol in each group.The result showed that the positive expressions of c-fos in paraventricular nucleus of hypothalamus both in sPCP and PCP groups were lower than that in vaccine group, and was lowest in sPCP low-dose group; Compared with vaccine group, the cortisol levels in sPCP low-dose group was significantly decreased in D21.The cortisol level in sPCP low-dose group and PCP high/low-dose group were lower than that in vaccine group, but there was no significant difference(P>0.05).The results confirmed that PCP and sPCP owned the effect of anti-immune stress, and with the best effect in low-dose of sPCP.
