阿魏菇粗多糖体外脾细胞调节功能及体内疫苗佐剂功能的研究

【摘要】 为研究阿魏菇粗多糖(PFCP)体外对小鼠脾细胞增殖的作用和体内对口蹄疫灭活疫苗的佐剂功能,MTT法检测PFCP最大无毒浓度,PFCP与Con A、LPS联用刺激小鼠脾细胞增殖的能力,ELISA检测细胞培养上清中IL-4与IFN-γ含量。PFCP联合口蹄疫病毒灭活抗原皮下注射免疫接种小鼠,ELISA检测IgG生成情况,流式细胞术分析免疫后小鼠脾脏T淋巴细胞亚群比例。结果显示,PFCP安全性好,体外可协同ConA与LPS促进小鼠脾细胞的增殖,增加IL-4与IFN-γ的分泌量。体内可增强口蹄疫病毒特异性IgG的水平,增加CD3~+CD4~+T、CD3~+CD8~+T淋巴细胞的数量。表明PFCP对小鼠脾细胞增殖具有较好的促进作用,且具有作为口蹄疫灭活疫苗佐剂的潜力。

【Abstract】 In order to study the effects of Pleurotus ferulae crude polysaccharide(PFCP) on the proliferation of mouse splenocytes in vitro and the adjuvant function of inactivated foot-and-mouth disease virus inactivated vaccine in vivo,MTT assay was used to detect the maximum non-toxic concentration of PFCP and the ability of PFCP combined with Con A and LPS to stimulate the proliferation of mouse splenocytes.IL-4 and IFN-γ contents in cell culture supernatant were detected by ELISA.Mice were immunized subcutaneously with PFCP combined with foot-and-mouth disease virus inactivated antigen.The production of specific antibody was detected by ELISA,and the proportion of spleen T lymphocyte subsets was analyzed by flow cytometry.The results showed that PFCP was safe and could cooperate with ConA and LPS to promote the proliferation of mouse splenocytes and increase IL-4 and IFN-γ secretions in vitro.It could enhance the expression of foot-and-mouth disease virus inactivated antigen-specific IgG and increase the number of CD3~+CD4~+T and CD3~+CD8~+T lymphocytes.The results showed that PFCP can promote the proliferation of splenocytes in mice, and has the potential to be used as adjuvant of inactivated foot-and-mouth disease vaccine.
