基于转录组学的中药药理机制分析方法及其在金花清感颗粒药理机制研究的应用
Analytical Method of Pharmacological Mechanism of TCM Based on Transcriptomics and Its Appication in Jinhua Qinggan Granules
【摘要】 目的:结合高通量转录组测序技术提出一种中药药理机制分析方法,并以金花清感颗粒为例探究其发挥药效作用的关键机制。方法:对中药作用于细胞系进行高通量转录组测序,获得中药作用转录组表达谱;设计一种基于基因共表达网络的生物信息学分析技术,即依次进行差异表达基因的识别、药效作用相似化合物的筛选、基因共表达网络的构建、基因模块的识别、GO富集分析、蛋白互作网络的构建、 HUB基因的识别、 HUB基因的验证。利用以上方法分析金花清感颗粒的药理机制。结果和结论:通过该方法发现金花清感颗粒对于新冠肺炎(COVID-19)患者具有治疗作用,且对病毒感染后血小板脱颗粒、血液高凝状态引起的多器官氧化应激损伤具有潜在的治疗修复作用,为进一步阐明金花清感颗粒的作用提供了证据。该方法适用于大多数中药药理机制的分析,为中医药现代化研究提供了方法学支撑。
【Abstract】 Objective:To propose a method combined with high-throughput transcriptome sequencing technology for analyzing the pharmacological mechanism of traditional Chinese medicine(TCM). Methods:By identification of differentially expressed genes, screening of compounds with similar pharmacodynamic effects, construction of gene co-expression networks;identification of gene modules, GO enrichment analysis, construction of protein interaction networks, identification of HUB genes, and verification of HUB genes, the pharmacological mechanism of jinhua qinggan granules was analyzed. Results and Conclusion:It is found that jinhua qinggan granules not only has therapeutic effects on COVID-19 patients, but also has potential therapeutic effects on multi-organ oxidative stress injury caused by platelet degranulation and blood hypercoagulability after virus infection. This method is suitable to analyze the pharmacological mechanism for most of TCMs,which provides methodological support for the modernization of TCM.
