响应面法优化蒙药白益母草总生物碱超声提取工艺

【摘要】 目的:建立蒙药白益母草中总生物碱的含量测定方法,考察蒙药白益母草总生物碱提取条件,以期为白益母草的总生物碱含量测定和质量控制提供实验依据。方法:以盐酸水苏碱为总生物碱含量测定的标准对照品,利用分光光度法测定白益母草中总生物碱含量,以总生物碱提取量为评价指标,选取乙醇浓度、料液比、提取时间为考察因素,采用单因素试验考察单因素最佳水平,采用Box-Behnken响应面法多指标综合考察最佳提取条件。结果:盐酸水苏碱在0~72 mg·L-1范围内具有良好的线性关系(r=0.999 9),回归方程为Y=-0.634 5X。白益母草总生物碱单因素考察表明70%乙醇提取、超声50 min、20倍(以上)料液比可获得最佳提取效果。响应面试验分析结果表明,乙醇浓度60%、料液比1∶15.52、提取时间50 min为白益母草总生物碱最佳提取条件,乙醇浓度与提取时间之间存在交互作用,乙醇浓度与料液比和料液比与提取时间之间无显著交互作用。结论:蒙药白益母草总生物碱的最佳提取工艺为:乙醇浓度60%、料液比1∶16、超声50 min。本提取方法操作简便,提取率高,可为白益母草的总生物碱含量测定和质量控制提供参考。

【Abstract】 Objective:To establish a method for the determination of total alkaloids in Mongolian herbal Baiyimucao, and to optimize the extraction conditions of total alkaloids in Baiyimucao, in order to provide experimental basis for the determination and quality control of total alkaloids in Baiyimucao(BAlk). Methods: Stachydrine hydrochloride was used as the standard control substance for the determination of total alkaloid content. The concentration of total alkaloid were determined by spectrophotometry. The effects of ultrasonic time,ethanol concentration and solid-liquid ratio on the extraction rate of total alkaloids were investigated. Based on the single factor tests, the Box-Behnken response surface methodology was used to optimize the extraction condition. Results: The content of stachydrine hydrochloride displays a good linear relationship(r= 0. 999 9) in the range of 0-72 mg·mL-1, and the regression equation wasY=-0. 6345 X. The single factor tests showed that 70% ethanol extraction, ultrasonic for 50 min, and the solid-liquid ratio 20 times(or above) could achieve the best extraction efficacy. The response surface design showed that the optimal process of ultrasonic extraction was as follow:ethanol concentration of 60%, solid-liquid ratio of 1∶ 15. 52, and extraction time of 50 min. There was an interaction between ethanol concentration and solid-liquid ratio. There was no interaction between ethanol concentration and solid-liquid ratio, and solid-liquid ratio and extraction time. Conclusion: The optimal extraction process of total alkaloids in the Mongolian herbal Baiyimucao is as follows: ethanol concentration of 60%, solid-liquid ratio of 1∶ 16, and ultrasound for 50 min. The extraction method has the advantages of simple operation and high extraction efficacy, and provides a reference for the determination of total alkaloid content and quality control of Baiyimucao.
