4株丁酸梭菌的抗逆特性体外评估

【摘要】 笔者通过模拟饲料制粒过程及家禽胃肠道环境,比较4株丁酸梭菌抗逆性能及生长能力的差异。选择4株丁酸梭菌(编号CB1~CB4)分别与基础饲粮混合制成试验日粮,试验日粮经模拟制粒温度、人工胃液、人工胰液、人工胆盐的耐受性以及丁酸产量等方面进行比较研究。结果显示,4株丁酸梭菌在模拟制粒温度下处理2.5 min,存活率均在30%以上,其中菌株CB1耐热性最佳,存活率为61.55%;耐人工胃液试验中,p H=2.0处理48 min后,菌株CB2的耐受性最好、CB1次之,存活率分别为74.07%和70.37%;耐消化道试验中,菌株CB1对人工胰液的耐受性最好,存活率达92.75%,在胆盐浓度为0.3g/L的环境中,菌株CB4对胆盐的耐受性最好,存活率达到58.82%。综上所述,将4株丁酸梭菌进行比较,菌株CB1的性能更优。本试验结果为4株丁酸梭菌的生产性能评价提供了参考依据。

【Abstract】 (objective) In this experiment, the stress resistance of 4 strains of Clostridium butyricum were compared by simulating the feed pelleting process and the gastrointestinal environment of poultry.(Methods) Four strains of Clostridium butyricum(no.CB1 to CB4)were mixed with the basal diet to prepare experimental diets, then the experimental diets were compared in terms of simulated granulation temperature, artificial gastric juice, artificial pancreatic juice, artificial bile salt tolerance and butyric acid yield.(Results) The results showed that the survival rates of the four strains of Clostridium butyricum treated at simulated granulation temperature for 2.5 min were more than 30%, of which strain CB1 had the best heat resistance and the survival rate was 61.55%; in the artificial gastric juice resistance test, after 48 min treatment with pH 2.0, the tolerance of strain CB2 was the best, followed by CB1, and the survival rates were74.07% and 70.37% respectively; in the digestive tract tolerance test, strain CB1 had the best tolerance to artificial pancreatic juice,with a survival rate of 92.75%; in the bile salt concentration of 0.3 g/L, strain CB4 had the best tolerance to bile salt, with a survival rate of 58.82%.(Conclusion) In conclusion, compared with the four strains of Clostridium butyricum, the performance of strain CB1 is better. The results of this test provide a reference basis for the evaluation of four Clostridium butyricum strains.
