赤松枯梢病生防真菌M75的筛选和鉴定

【摘要】 为筛选对松枯梢病原菌具有拮抗作用的菌株,以赤松枯梢病病原菌松球壳孢菌(Sphaeropsis sapinea)为供试病原菌,赤松林地土壤真菌为材料,采用平板对峙法和菌丝生长速率法进行拮抗真菌筛选,获得对松枯梢病原菌有显著抑制效果的土壤真菌M75。采用菌丝生长速率法测定所获拮抗真菌对金黄壳囊孢(Cytospora chrysosperma)等8种常见真菌的抑菌效果。结果表明:拮抗真菌M75平板对峙抑菌率为72.65%,培养液复筛抑菌率为78.78%,经形态学及分子生物学将该真菌鉴定为森吉木霉(Trichoderma songyi)。森吉木霉对8种常见真菌均具有一定的抑菌效果,对金黄壳囊孢抑制率为94.05%;对葡萄座腔菌(Botryosphaeria dothidea)抑制率为82.30%;对栗疫菌(Cryphonectria parasitica)抑制率为77.87%。菌株M75抑菌谱广,在生物防治领域具有较大应用潜力。

【Abstract】 In order to screen the strains with antagonistic effect on the pathogen of pine shoot blight, the soil fungus M75 with significant inhibitory effect on the pathogen of pine shoot blight was obtained from the forest soil fungi of Pinus densifloraby using the plate confrontation method and mycelial growth rate method. The antibacterial effect of the obtained antagonistic fungi on 8 common fungi including Cytospora chrysosperma was determined by the mycelial growth rate method. The plate confrontation inhibition rate of antagonistic fungus M75 was 72.65%, and the rescreening inhibition rate of fermentation broth was 78.78%. Through morphological and the tef1-a gene sequence analysis, the strain M75 was identified as Trichoderma songyi. The strain has a broad spectrum of antimicrobial activity against 8 common fungi, the inhibition rate of the C. chrysosperma was 94.05%, the inhibition rate of Botryosphaeria dothidea was 82.30% and the inhibition rate of C. parasitica was 77.87%. The strain M75 has a broad antibacterial spectrum and has great application potential in the field of biological control.
