超声波辅助提取泡桐花总黄酮工艺及抑菌活性研究
Study on ultrasonic-assisted extraction process and antibacterial activity of Flos Paulowniae Fortunei total flavonoids
【摘要】 为探究超声辅助提取泡桐花总黄酮最优工艺条件,以泡桐花总黄酮含量为测定指标,通过单因素和响应面试验优化提取工艺条件。结果表明,超声波辅助提取泡桐花总黄酮最佳提取工艺条件为:乙醇浓度75%,超声时间66 min,超声功率310 W,温度61℃,得率7.49%,与模型预测值7.53%接近。总黄酮体外抑菌试验、最小抑菌浓度(MIC)和最小杀菌浓度(MBC)测定结果表明,泡桐花总黄酮对金黄色葡萄球菌、链球菌、沙门氏杆菌和大肠埃希菌均有不同程度抑菌活性,MIC分别为0.39、1.57、50.00和12.50 mg·mL-1,MBC分别为0.78、3.13、50.00和25.00 mg·mL-1。泡桐花总黄酮在100~0.39 mg·mL-1浓度对金黄色葡萄球菌抑菌效果显著(P<0.05)。综上,泡桐花总黄酮应用超声波辅助提取工艺具有可行性,具有一定体外抑菌作用,其中对金黄色葡萄球菌抑菌效果最佳。
【Abstract】 In order to explore the conditions for extracting the total flavonoids from Flos Paulowniae Fortunei by ultrasonic-assisted method, the contents of total flavonoids rate were set as the index, and extraction process was optimized by single factor test and response surface methodology. The results indicated that the optimum extraction conditions of the total flavonoids from Flos Paulowniae Fortunei were as the following: extraction temperature 61 ℃, ultrasonic power 310 W, ultrasonic time 66 min,and ethanol concentration 75%, the actual yield of total flavonoids was 7.49%. It was basically consistent with the predicted value(7.53%). According to the result of in vitro antibacterial test, minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC), Flos Paulowniae Fortunei total flavonoids showed antibacterial activity against Staphylo-coccus aureus, Streptococcus, Salmonella and Escherichia coli Staphylococcus aureus. The MIC of Flos Paulowniae Fortunei total flavonoids against Staphylococcus aureus, Streptococcus, Salmonella and Escherichia coli were 0.39, 1.57, 50.00 and12.50 mg · mL-1, respectively. The MBC were 0.78, 3.13, 50.00 and 25.00 mg · mL-1, respectively. There was a significantly antibacterial effect against Staphylococcus aureus, when Flos Paulowniae Fortunei total flavonoids concentration at 100-0.39 mg · mL-1(P<0.05). In conclusion, the result indicated that it was feasible to extract total flavonoids from Flos Paulowniae Fortunei by ultrasonic-assisted extraction method. The total flavonoids from Flos Paulowniae Fortunei showed antibacterial activity, especially against Staphylococcus aureus.
